酸性神经肽1对小鼠大脑氨基酸类神经rn递质及cGMP影响的实验研究

         

摘要

Objective: The aim of the research is to probe the changes of some neurotransmitters of amino acid (glutamic acid,Glu)and (γ-aminobutyric acid,GABA)and cGMP in mice brains caused by bovine acidic neuropeptide 1(BANP-1).Methods: Mice were separated into six groups (n=15 at each group).Five of the groups were injected with BANP 1 into mice's abdominal cavities at dose of 15 mg/kg and brain was collected and homogenated at 0.5 h,1 h,2 h,3 h and 4 h for examination.The other group was injected with normal saline as control group and brain was collected at 1 h for examination.Brain Glu and GABA were analysised by filter paper-electrophoresis method.Brain cGMP were analysised by radiation immunity method.Results: After injection of BANP-1 in 1 h, 2 h,3 h,4 h,the results were compared with that of control group.It indicated that:Glu concentration had a significant increase in 1 h,2 h(P<0.01,P<0.01 respectively).GABA also increased in 2 h,3 h(P<0.01 respectively).cGMP concentration increased in 1 h,2 h(P<0.05,P<0.01 respectively).Conclusion: After injection of BANP-1,the concentrations of Glu and GABA and cGMP increased at different time.%目的: 探讨酸性神经肽(bovine acidic neuropeptide 1 BANP-1)对小鼠脑内氨基酸类递质(谷氨酸和γ-氨基丁酸)及cGMP的影响。方法: 将小鼠随机分为6组(每组15只),其中5组按15 mg/kg腹腔注射BANP-1,将小鼠在注射后0.5 h,1 h,2 h,3 h,4 h处死并取脑匀浆,另一组为注射生理盐水对照组,在注射盐水后1 h处死并取脑匀浆,用75%冰乙醇抽提氨基酸类物质及cGMP并沉淀蛋白质。应用滤纸电泳法对小鼠大脑谷氨酸(glutamic acid,Glu)和γ-氨基丁酸(γ-aminobutyric acid,GABA)进行测定,应用放射免疫法对小鼠大脑环磷酸鸟苷(cyclic monophosphate guanosine,cGMP)进行测定。结果: 腹腔注射BANP-1后,各项指标在不同时间的结果分别与对照组比较:Glu浓度在1 h、2 h时明显高于对照组(P<0.01),0.5 h、3 h、4 h时差异无显著性(P>0.05)。GABA浓度在2 h、3 h时显著高于对照组(P<0.01),0.5 h、1 h、4 h时差异无显著性(P>0.01)。cGMP浓度有一定升高,1 h 2 h时显著升高(P<0.01),其余各时间组差异无显著性(P>0.05)。结论: BANP-1急性用药后在一定时间可升高脑内Glu、GABA及cGMP水平。
