目的：探讨姜黄素对缺血性脑损伤的神经保护作用及机制。方法雄性健康昆明小鼠，随机分为A组：假手术组；B组：缺血再灌组；C组：姜黄素治疗组。3组于缺血再灌注后分为30 min、3 h、12 h、24 h、48 h、72 h等6个亚组。采用线栓法建立小鼠局灶性脑缺血再灌注模型。 TTC染色法测定脑梗死体积，水迷宫法测试动物学习记忆功能；免疫组织化学法观察caspase-8蛋白的表达及分布；免疫印迹法检测皮质区caspase-8蛋白表达量；利用原位缺口末端标记法（ TUNEL法）研究神经细胞凋亡的变化。结果24 h脑梗死体积B组高于A、C组（ P ＜0â．05）。 B组第9、10天搜索安全岛潜伏期较A、C组明显延长（ P ＜0．01）。与A组比较，B组caspase-8在再灌后3 h、12 h、24 h、48 h显著增高（ P ＜0．05），与B组相比C组显著回降（ P ＜0．05）。 C组与B组比较，各时相点凋亡细胞显著减少（ P ＜0．05）。结论姜黄素的神经保护机制之一为减少caspase-8的表达，从而减轻细胞凋亡而发挥神经保护作用。%Objective To investigate curcumin ’ s neuroprotective effect and its mechanism in mice with focal cerebral ischemia reperfusion .Methods Healthy male Kunming mice were randomly divided into three groups: group A ( sham-operated group),group B (ischemia reperfusion group),group C (curcumin treatment group);all of them were designed to 6 subgroups (30min,3h,12h,24h,48h and 72h).Morris water maze test was used to detect the leaning and memory function . Immunohistochemistry and Western Blotting were used to detect the caspase-8 expression .Results Infarct volume of group B at 24h was significantly higher than that in group A ( P <0.05),while that in group C was apparently decreased ( P <0.05).Morris water maze test showed:For the searching refuges latency in 9 and 10 days,group B was significantly prolonged than group A , while group C was significantly shorter than group B .Immunohistochemistry and Western-blot showed: After ischemia reperfusion,caspase-8 of group B was significantly higher than group A in 3h,12h,24h,48h( P <0.05); group C was significantly back down compared with group B in 3h,12h,24h( P <0.05).Apoptotic cells of group C were obviously decreased than group B ( P <0.05 ).Conclusion Curcumin has protective function on mice focal cerebral ischemia reperfusion by lowering the caspase-8 expression to decrease cell apoptosis and improving leaning and memory function .
