龙胆水提物对急性胰腺炎大鼠肝损伤的保护作用及机制研究

         

摘要

Objective To investigate the protective effect and action mechanism of water extract of gentian on liver damage of rats with acute pancreatitis (AP).Methods Forty SD rats were randomly divided into normal control group ( n =8) and model group ( n =32).The animal models with AP were established by injecting 5% cow-bezoar sodium cholate through the duodeno papillary retrograde cholangiopancreatic duct.After modeling,the rats were redivided into four subgroups :model control group,low-dose group (water extract of gentian 12mg/kg),median-dose group (24mg/kg) and high-dose group (48mg/kg),with 8 rats in each group.The rats in low-dose,median-dose and high-dose groups were given water extract of gentiane 12mg/kg,24 mg/kg,48 mg/kg,respectively by gavage,however,the rats in normal control group and model control group were given equal volume of 0.9% sodium chloride solution,once a day for 1 week.After administration,the serum levels of ALT, AST, AMY, IL-6 and TNF-α were detected.Moreover the rats were sacrificed and liver specimens were taken out,the changes of liver structure were observed by HE staining,and the expression levels of NF-κB and I-κB in liver tissue were detected by Western Blot.Results As compared with those in model control group, the serum levels of ALT,AST,AMY, IL-6,TNF-α in median-dose group and high-dose group were significantly decreased ( P <0.01),and the expression levels of NF-κβprotein were obviously decreased too ( P <0.01),however, the expression levels of I-κB protein were significantly increased ( P <0.01).Conclusion The water extract of gentian can obviously improve the liver damage caused by acute pancreatitis in rats.%目的 探讨龙胆水提物对急性胰腺炎(AP)大鼠肝损伤的保护作用及机制.方法 SD大鼠40只随机分为正常组(n=8)与模型组(n=32),模型组采用经十二指肠乳头逆行胆胰管注射5%牛黄胆酸钠方法制备AP大鼠模型,造模后大鼠分为4个亚组:模型对照组(n=8)、龙胆水提物12 mg/kg组(n=8)、24 mg/kg组(n=8)、48 mg/kg组(n=8),龙胆水提物低、中、高剂量组每天分别灌胃给予龙胆水提物12 mg/kg,24 mg/kg及48 mg/kg,正常组和模型对照组给予等体积的0.9%氯化钠溶液,连续给药1周,1次/d.给药结束后颈总动脉取血,分离血清,检测血清ALT、AST、AMY、IL-6、TNF-α水平,大鼠脱颈椎处死后剪取肝右叶1/2,于4%中性甲醛固定,HE染色观察肝脏结构变化,western blot 检测肝脏组织NF-κβ及I-κB表达水平.结果 与模型对照组比较,龙胆水提物24 mg/kg及48 mg/kg组血清ALT、AST、AMY水平及IL-6及TNF-α表达水平显著降低(P<0.01);肝组织NF-κβ蛋白表达水平显著降低(P<0.01),I-κB蛋白表达水平显著升高(P<0.01).结论 龙胆水提物能够显著改善AP导致的肝损伤.
