甲型肝炎病毒单克隆抗体细胞株的建立及鉴定

摘要

Objective To discuss the establishment and identification of monoclonal antibody (McAb) cell lines against hepatitis A virus (HAV). Methods The McAb cell lines against HAV were cultured. The stable McAb hybridoma cell lines were selected and numbered with A, B, C, which was then detected for specificity and titer. Results The titer of McAb hybridoma cell lines and HAV antigen was 1: (10000-80000). The titer of McAb cell culture supernatant and HAV antigen was 1:(2048-16824). Conclusion The McAb hybridoma cell lines has good stability and specification, as well as high titer, which is of great important to the clinical diagnosis of HAV and the vaccine culture inoculation.%目的 探讨建立和鉴定甲型肝炎(HAV)病毒单克隆抗体(McAb)细胞株的方法,为临床提供抗体来源.方法 通过McAb杂交瘤细胞培养方法进行甲型肝炎病毒单克隆抗体细胞株的培养后,选取实验培养的具有稳定性的McAb杂交瘤细胞后编号,对各细胞株进行特异性、效价的测定,并进行比对.结果 McAb杂交瘤细胞与甲肝病毒抗原的结合特异性效价在1:( 10 000～80 000)间,提示McAb与甲肝病毒抗原的结合具有特异性；McAb细胞培养上清液效价在1:(2 048～16 824)间.结论 通过McAb杂交瘤方法建立的甲型肝炎病毒单克隆抗体细胞株具有较好的稳定性和特异性,且其效价较高,对临床甲型肝炎的诊断和疫苗的培养接种具有十分重要的作用.
