1株碱蓬内生芽孢杆菌的分离及抑菌活性测定

     

摘要

The morphology, colonial feature, growth characteristics and bacteriostatic activity of an endophytic bacillus strain (EB0901) isolated from healthy tissue of Suaeda glauca were studied to probe the biological control mechanism of endophytic bacteria. The results showed that the optimum temperature, pH and NaCl concentration for EB0901 growth was 35℃, 7 and 0.5%, EB0901 had certain inhibition effect on mycelium growth of 9 tested plant pathogenic fungi, the bacteriostatic rate of EB0901 against Exsserohilum turcicum , Sc ferotiniasc ferotiorm , Bipolarisi and Gloeosporium piperatum was above 60%, significantly higher than other five tested plant pathogenic fungi. The bacteriostatic rate of EB0901 against Exserohilum turcicum reached 64. 11%, which indicated that EB0901 with strong bacteriostatic activity should be developed and utilized further.%为进一步探究内生细菌的生防机制,采用NA固体倾注法培养从碱蓬健康组织中分离的菌株,并对分离菌株的个体形态、菌落特征和生长特性进行初步研究,同时采用平板对峙法对其抑菌活性进行了测定.结果表明:从碱蓬健康组织中分离出1株内生芽孢杆菌(EB0901);该菌的生长最适温度为35℃,初始pH6~8时出现生长高峰,以pH为7时生长最好;在NaCl浓度为0.5%时生长最佳,但在10%以上时仍可生长;对供试的9种植物病原真菌菌丝生长均有一定的抑制作用,对玉米大斑病菌、辣椒菌核病菌、平脐孺孢病菌、辣椒炭疽病菌的抑菌能力较强,抑菌率在60%以上,显著高于其他5种植物病原真菌(P<0.05),其中,对玉米大斑病菌的抑菌活性最强,抑菌率高达64.11%.碱蓬内生芽孢杆菌是一种具有明显抑菌活性的内生细菌,值得进一步开发利用.
