马铃薯转三价病毒 YRX 外壳蛋白基因片段株系的核型及荧光原位杂交分析

摘要

The chromosome of transgene potato Shepody line pHeYCPC326-XC-Sh was analyzed by karyotype analysis and FISH to study the location of exogenous gene YRX (the conserved fragment of potato virus X,potato virus Y and potato leaf roll virus)on chromosome of transgenic potato and its genetic stability. Shepody is an autotetraploid with 48 chromosomes with an arm ratio of (1.37±0.23)~(2.28±0.14).The karyotype of Shepody is 2B and the karyotype formula is 2n=4x=7m+ 5sm.There are satellite chromosomes in chromosome C and J chromosome.The exogenous gene YRX on chromosome A,B,E,G and I is located in the telos of the chromosomes’short arm.%为了解外源三价外壳蛋白基因保守片段重组序列 YRX（马铃薯 X 病毒、Y 病毒和卷叶病毒）在转基因马铃薯染色体中的位置，分析其遗传稳定性，采用核型分析与荧光原位杂交技术对转基因马铃薯夏波蒂株系 pHeYCPC326-XC-Sh 进行染色体分析。结果表明：夏波蒂的染色体数目为48条，臂比范围为（1．37±0．23）～（2．28±0．14），核不对称属2B 型，核型公式为2n＝4x＝7m ＋5sm，其中 C 组与 J 组含随体。发现外源基因 YRX 在染色体 A、B、E、G、I 组均存在，大多位于染色体短臂端部。
