红花多糖对人胃癌SGC-7901细胞抑制作用的初步研究

摘要

目的 观察红花多糖(SPS)对人胃癌SGC-7901细胞增殖与凋亡的影响.方法 不同浓度的SPS相同时间作用于人胃癌SGC-7901细胞,采用倒置显微镜观察SGC-7901细胞的形态学变化及增殖情况,吖啶橙染色观察SGC-7901细胞的凋亡情况.结果 SPS处理组贴壁细胞的数量明显减少,细胞体积变小,部分细胞崩解.吖啶橙染色结果 显示,经SPS处理组细胞胞质浓缩、染色增强,部分细胞核固缩为新月状.结论 SPS能抑制人胃癌SGC-7901细胞体外生长,同时可诱导SGC-7901细胞凋亡,该作用呈一定的时间与剂量依赖.%Objective To observe the effeet of Safftower potysaccharide( SPS ) on the eett protiferation and apoptosis of human gastric eareinoma eett tine SGC-7901. Methods SGC-7901 eetts were proeessed with different. eoneentrations of SPS during the same period. The morphotogy ehanges and pmtiferation of SGC-7901 eetts were observed wiht inverted miemseope. The apoplosis of SGC-7901 cott was detected by acridine orange(AO ) assay. Resutts SGC-7901 eetts in SPS group showed bad refraction,shrink,and more dead eetts in culture medium were observed. Resulls of AO ftuoreseent staining showed part of SGC-7901 eetts of eytoptasm condensed,chromatin increased,and the crescent-shaped shrinkage coutd be seen in some cetts nucteus. Conctusion SPS can markedty inhibit. the growth of SGC-7901 cetts and induce apoptosis of gastrie carcinoma cett tine SGC-7901 in vitro cukured in a time- and dose-dependent manner.
