高压氧联合抗震颗粒与美多芭对帕金森病大鼠黑质Bcl-2、Bax影响的对比研究

             

摘要

Objective To compare the effects of hyperbaric oxygen combined with anti-parkinson granule,levodopa and benserazide hydrochloride tablets on the expression of Bcl-2 and Bax protein in experimental parkinsonian rats. Methods The parkinsonian rat model was established with injection of 6-hydroxydopamine( 6-OHDA ) into the ventral tegmental of right side brain. The 18 model rats were randomly divided into 3 groups: the treatment group by using hyperbaric oxygen combined with anti-parkinson granule( n =6 ),the treatment group by using levodopa and benserazide hydrochloride tablets( n =6 ) and the model group( n =6 ). After the subjects were treated for 14 days,expression of Bcl-2 and Bax protein in parkinsonian rats were detected by immunohistochemistry assay, and the changes of ethology were observed in the 3 groups. Results No significant difference was found in the positive expressions of Bcl-2 and Bax protein between the two treatment groups( P >0.05 ). The positive expressions of Bcl-2 were significantly increased,and the positive expressions of Bax protein were decreased in the two treatment groups compared with those in the model groups( P <0.05 ). Conclusion Hyperbaric oxygen combined with anti-parkinson granule,can significantly restrain apoptosis of substantia nigra as well as levodopa and benserazide hydrochloride tablets, and shows a good curative effect in the treatment of parkinsonian rats.%目的 对比研究高压氧联合抗震颗粒与美多芭对实验性帕金森病大鼠黑质Bcl-2、Bax表达的影响及意义.方法 采用单点注射6-羟基多巴胺(6-OHDA)建立偏侧帕金森病大鼠模型18只,随机分为高压氧联合抗震颗粒组6只,美多芭组6只,模型组6只,给药后14 d,采用免疫组织化学法检测各组大鼠大脑黑质Bcl-2、Bax的表达,并观察各组大鼠治疗前后有无异常行为.结果 高压氧联合抗震颗粒组与美多芭组的Bcl-2、Bax蛋白阳性表达率比较差异均无统计学意义(P>0.05);高压氧联合抗震颗粒组及美多芭组Bcl-2蛋白阳性表达率明显高于模型组,Bax蛋白阳性表达率明显低于模型组(P<0.05).结论 高压氧联合抗震颗粒与美多芭均能抑制帕金森病大鼠黑质细胞凋亡,对帕金森病大鼠有治疗作用.
