急性ST段抬高心肌梗死患者LDLR基因启动子区的甲基化修饰

         

摘要

目的 分析急性ST段抬高心肌梗死(STEMI)患者低密度脂蛋白受体(LDLR)基因启动子区CpG岛甲基化修饰程度.方法 选择经急诊确诊的STEMI患者48例(STEMI组),立即抽取静脉血查血脂和保存用于DNA提取;以健康人群36例为对照组,亚硫酸氢盐修饰后测序检测其LDLR基因启动子区甲基化程度.结果 STEMI组患者血清低密度脂蛋白(LDL)水平较对照组升高,在LDLR基因启动子区CpG岛(-383,-140)片段中所包含的7个CG位点均无发生甲基化.结论 STEMI患者血清LDL水平改变与LDLR基因启动子区CpG岛(-383,-140)的甲基化修饰无关.%Objective To analyze the methylation status of CpG island in low density lipoprotein receptor ( LD-LR ) gene promoter in patients with acute ST segment elevation myocardial infarction ( STEMI ). Methods Forty - eight STEMI patients and 36 healthy controls were recruited between December 2009 and June 2011. Peripheral blood was taken for DNA extracted and lipid assessment. Methylation of CpG island in the promoter of LDLR gene was determined by Bisulfite Sequence Polymerase chain reaction ( BSP ). Results The serum LDL of STEMI patients were significantly higher than that of healthy controls. No methylation was detected in the CpG island fragment ( - 383 , - 140 ) of LDLR gene promoter. Conclusion The elevation of serum LDL is not associated with the methylation CpG island fragment (-383,-140 ) of LDLR gene promoter in STEMI patients.
