健心颗粒对慢性心衰大鼠心室肌 toll样受体4、肿瘤坏死因子-α蛋白表达的影响

         

摘要

目的:探讨健心颗粒对慢性心衰大鼠心室肌toll样受体(toll-like receptors, TLR)4、肿瘤坏死因子( tumor necrosis factor, TNF)-α蛋白表达的影响。方法将15周龄的自发性高血压大鼠120只随机分为培哚普利组、健心颗粒低剂量组、健心颗粒中剂量组、健心颗粒高剂量组、阴性对照组,每组24只。前四组分别予培哚普利0.36 mg/( kg·d),健心颗粒1.6 g/( kg·d)、3.2 g/( kg·d)、6.1 g/( kg·d),以2 mL生理盐水溶解,阴性对照组予生理盐水2 mL,灌胃给药,每天2次。实验后每7天各组分别随机处死6只大鼠,采用免疫印迹检测上述各点心室肌组织TLR4蛋白表达,免疫组织化学显色检测上述各点心室肌组织TNF-а蛋白表达。结果第7天,对照组、健心颗粒低、中、高剂量组间比较,大鼠心室肌 TLR4、TNF-α蛋白表达无显著差异( P>0.05 ),但培哚普利组明显下降(P<0.05);第14天、21天,与对照组比较,其他各组大鼠心室肌TLR4、TNF-α蛋白表达均显著下降(P>0.05),但培哚普利组较其他组下降更明显(P<0.05);第28天,与对照组比较,其他各组大鼠心室肌TLR4、TNF-α蛋白表达均显著下降(P>0.05),但培哚普利组、健心颗粒中、高剂量组明显低于低剂量组(P<0.05)。结论健心颗粒具有显著抗炎作用。%Objective To investigate the effect of the Jianxin Granule ( JG) in the expression of TLR4 (toll-like receptors 4) and TNF-αon ventricular muscle in rats with chronic heart failure. Methods rats ( SHR-sp) 120 rats, which are 15-week-old with spontaneous hypertension, were randomly divided into five groups:perindopril group 0. 36 mg/(kg·d), low dose JG group 1. 6 g/(kg·d), median dose JG group 3. 2 g/(kg·d), high dose JG group 6. 1 g/(kg·d), and negative control group (with saline 2 ml) . There are 24 rats for each group. The drugs, dissolved in 2 ml saline water, were administrated by ig twice a day. During the experiment (28 days), 6 rats in each group were randomly executed every 7 days. Meanwhile the expression of TLR was determined by Western blot and pro-TNF-αby immunohistochemical staining. Result On the seventh day, there were no significant difference (P>0. 05) of the expression of TLR4 and TNF-α in ventricular muscle among the control group, low, medium and high dose groups, but the perindopril group decreased obviously (P<0. 05). On the fourteenth and the twenty-first day, com-pared with the control group, the expression of TLR4 and TNF-αin ventricular muscle groups significantly decreased (P >0. 05), furthermore, the perindopril group decreased more significantly than the other groups. On the twenty-eighth day, the expression of TLR4 and TNF-α in ventricular muscle groups de-creased more significantly than the control group (P>0. 05), however, the perindopril group, medium and high dose groups were significantly lower than those in the low dose group (P<0. 05). Conclusion Jiax-in granule has a significant anti-inflammatory effect.
