运用RP-HPLC测定油茶枯饼中皂苷类、黄酮类有效指标性成分的含量

摘要

Objective To establish a HPLC method for the determination of index components of flavonoids and saponins in oil-tea camellia defatted cake based on drug system.Methods Reversed phase high performance liquid chromatography was used for determination of index components of flavonoids and saponins in acid system at the same time, Waters×bridge C18 (4.6×250mm, 5μm) chromatographic column was used;moving phase was acetonitrile and 0.05% phosphoric acid water,gradient elution,flow rate was 1 mL/min, column temperature was 30℃.The detection wavelength of flavonoids was 265 nm.The detection wavelength of saponins was 203nm, the injection volume was 20μL.Results The kaempferol 3-O-[2-O-β-D-xylose-6-O-α-L-rhamnopyranosyl]-β-D-glucopyranoside, kaempferol3-O-[2-O-β-D-galactosel-6-O-α-L-rhamnopyranosyl]-β-D-glucopyranoside and camelliasaponin B1 showed a good linear relationship with peak area respectively in 0.0876~8.76 mg/mL, 0.0885~8.85 mg/mL, 0.0872~8.72 mg/mL, and the correlation coefficient was 0.9999.Precision, stability and repeatability were reached the requirements.The average recoveries were 99.22%, 101.50%, 100.44%, and RSD were 1.66%, 1.72%, 1.58%, respectively.Conclusion The method is quick, simple, accurate and reproducible, and can be used for the quality control and content determination of oil-tea camellia defatted cake.%目的 基于药物体系导向建立油茶枯饼中皂苷类和黄酮类有效指标性成分的含量测定方法. 方法 采用反相高效液相色谱方法,在酸性系统下同时测定油茶枯饼中皂苷类和黄酮类有效指标性成分,采用Waters×bridge C18(4.6×250 mm,5 μm)色谱柱;流动相为乙腈-0.05%磷酸水,梯度洗脱,流速1.0 mL/min,柱温30℃,黄酮类检测波长为265 nm,皂苷类检测波长为203 nm,进样20 μL.结果 山奈酚3-O-[2-O-β-D-木糖-6-O-α-L-鼠李糖]-β-D-葡萄糖苷、山奈酚3-O-[2-O-β-D-半乳糖-6-O-α-L-鼠李糖]-β-D-葡萄糖苷、camelliasaponin B1分别在0.0876~8.76 μg、0.0885~8.85 μg、0.0872~8.72 μg范围内与色谱峰面积呈现良好的线性关系,相关系数均为0.9999.精密度、稳定性、重复性良好,平均加样回收率分别为99.22%、101.50%、100.44%,RSD值分别为1.66%、1.72%、1.58%. 结论 所建方法快速、简便、准确度高、重现性好、适用于油茶枯饼的含量测定与质量控制.
