枸杞多糖对HepG2细胞膜表面超微结构变化的影响

         

摘要

目的:研究枸杞多糖对人肝癌HepG2细胞膜表面超微超微结构变化的影响.方法:利用倒置显微镜和原子力显微镜观察不同浓度(0、50、100、200、400μg/mL)和不同作用时间下(24h、48h)枸杞多糖对HepG2细胞膜表面结构变化的影响.结果:随作用时间和多糖浓度的增加,细胞膜表面形态发生了明显的变化,膜表面结构由光滑变得粗糙,细胞膜表面粗糙度由1.451nm变到295.977nm,部分形成孔洞和隆起,表面结构遭到了明显的破坏.结论:枸杞多糖能破坏HepG2细胞膜结构,这可能与枸杞多糖的抗肿瘤作用有关.%[Objectect] To research the effect of Lycium barbarum polysaccharides on the ultrastructure of membrane surface of human hepatic HepG2.[Method] To study the impact of the Lycium barbarum polysaccharides on the HepG2 cells and the ultrastructural changes of cell surface with an inverted microscope and atomic force microscopy at different concentrations (0,50,100,200,400μg/mL) and different time (24h,48h).[Result] With the gradual increase of time and polysaccharides concentration,the membrane surfaces of the cells began to change significantly.The cell surface gradually became uneven,cell roughness changed from 1.451nm to 295.977nm,and part of the cell surface formated voids and uplift.The surface of the membrane structure suffered significant damage.[Conclusion] The Lycium barbarum polysaccharides could destroy the structure of the HepG2 cell membrane,which might be related to the anti-tumor effect of the Lycium barbarum polysaccharides.
