ROL酶解制备富含甘油二酯的大豆油工艺的优化

     

摘要

以大豆油为试验原料,采用3种不同的脂肪酶(RML,ROL和ALIP)催化水解产生甘油二酯,并对其生产条件进行优化.结果表明:ROL酶催化水解产生的1,3-二亚油酸甘油酯的含量最高,为16.78%;进一步研究不同反应时间(1~9 h),温度(25~60℃),加酶量(12~128 U/g,占油重),加水量(28%~80%,占反应体系总重),搅拌方式和搅拌速度(50~900 r/min)对ROL水解大豆油产生甘油二酯含量的影响.在2.5 g的大豆油中加64 U/g ROL酶液,反应体系中含水量为28.57%,温度为30℃,磁力搅拌器转速为500 r/min,反应时间为4h的条件下,酶解产物中1,3-二亚油酸甘油酯的含量最高,达23.91%,产物中总的甘油二酯含量约为55.5%.%Three lipases (RML, ROL, and ALIP) were used for producing diacylglycerols (DAGs) from soybean oil and optimized the reaction conditions. The results showed that, ROL had the highest ability for producing 1, 3-dilinolein with the highest yield of 16.78 wt %. Subsequently, ROL was selected for evaluating various conditions on the effect of 1, 3-dilinolein yield, which including reaction time {1~9 h), reaction temperature (25-60℃), enzyme loading (12-128 U/g of oil), wa-ter addition (28%~80%WT of total reaction system), stirring method and mixing speed (50-900 r/min). The optimal conditions were obtained and shown as follow: reaction temperature 30℃, enzyme loading 64 U/g, water content 28.57%, reaction time 4 h. Under these conditions, the yield of 1, 3-dilinolein was 23.91%, and the total content of diacyl-glycerol was about 55.5%.
