短小芽孢杆菌产角蛋白酶液体发酵条件的优化

         

摘要

In order to enhance the Keratinase activity of Bacillus pumilus strain ZW-36, the influence of initial pH, concentration of feathers, inorganic salt, external carbon source, external nitrogen source on Keratinase production were studied. Designed with single factor experiment and the Box-Behnken in the response surface methodology, a quadratic regression model of ferrous sulfate, magnesium sulfate and dipotassium phosphate was established. The optimization conditions of producing Keratinase were initial pH at 6.5, 0.5%of chicken feather, 0.5% of sodium chloride, 0.036% of ferrous sulfate, 0.025% of magnesium sulfate, 0.073%of dipotassium phosphate, 0.035% of potassium dihydrogen phosphate and 0.5%of corn flour. Under this conditions, the Keratinase activity in fermentation broth of shask on the 36 hours was 57.14U/mL, which was higher 49.3% than before.%为提高Bacillus pumilus ZW-36产角蛋白降解酶能力,考察了初始pH、羽毛浓度、无机盐、外加碳氮源等因数对其产角蛋白酶的影响。采用单因素试验和响应面分析法的Box-Behnken进行试验设计,建立了FeSO4、MgSO4和K2HPO4的二次回归模型。得到产角蛋白酶的最佳条件为:培养基初始pH 6.5、羽毛0.5%、NaCl 0.5%、FeSO40.036%、MgSO40.025%、K2HPO40.073%、KH2PO40.035%,玉米粉0.5%。在此条件下发酵36h角蛋白酶活达到57.14U/mL,达到模型预测值的96.7%,较优化前(38.27U/mL)酶活提高了49.3%。
