目的 用分子生物学方法,对僵蚕进行初步鉴定研究,研究其来源昆虫 COI(cytochrome oxidase subunit I,细胞色素氧化酶亚基 I)和来源真菌 ITS(Internal Transcribed Sequence, 内转录间隔区)区的基因序列.方法 对来源昆虫 COI 和来源真菌 ITS2 区基因进行了 PCR 扩增和序列测定,比对分析序列,并与 GenBank 核酸序列数据库中的序列进行检索比对.结果 发现多批次僵蚕来源昆虫的COI 基因序列高度一致,但来源真菌ITS 基因呈现多样性.结论 可通过真菌ITS序列结合来源昆虫 COI序列鉴定僵蚕的真伪.%Objective To identify medicinal material Bombyx batryticatus and its counterfeits by a gene sequence study of the rDNA ITS (internal transcribed sequence) region of fungi and COI (cytochrome oxidase subunit I) gene of host insects. Methods The COI gene of host insects and rDNA ITS region of fungi were amplified and sequenced. The sequences were analyzed and compared with the sequences in Genbank database. Results For the sequence of COI gene of host insects from all the collected samples, there was high similarity, while the rDNA ITS region of fungi had genetic diversity. Conclusion The combination of ITS region of fungi and COI gene of host insects can be used to identify Bombyx batryticatus and its adulterants effectively.
