白藜芦醇对膀胱癌耐药细胞株BIU-87/ADM自噬影响的实验研究

             

摘要

目的:探讨白藜芦醇对膀胱癌耐药细胞株BIU-87/ADM自噬的影响.方法:分别建立浓度为12.5μmol/L、25.0μmol/L、50.0μmol/L与100μmol/L的白藜芦醇制备品,并以未加药人膀胱癌BIU-87/ADM细胞作为对照.通过CCK-8法检测对照组及不同浓度梯度白藜芦醇对膀胱癌耐药细胞株BIU-87/ADM增殖的影响,应用RT-PCR法测定白藜芦醇处理前后膀胱癌耐药细胞株BIU-87/ADM LC-3和Beclin 1 mRNA的表达水平,并以Wester blot检测白藜芦醇作用后LC-3Ⅱ蛋白的表达水平.结果:加入白藜芦醇试验组BIU-87/ADM细胞24h与48h生长抑制率均明显高于对照组,其48h生长抑制率明显高于24h生长抑制率,具有统计学意义(P<0.05),白藜芦醇浓度越高其细胞生长抑制率越高.加入白藜芦醇试验组BIU-87/ADM细胞的细胞凋亡率明显高于对照组,具有统计学意义(P<0.05),白藜芦醇浓度越高其细胞凋亡率越高.白藜芦醇干预后LC3-Ⅱ与Beclin 1 mRNA表达明显减弱,而白藜芦醇浓度越高其表达减弱程度越明显.结论:白藜芦醇能够明显减少耐药细胞株BIU-87/ADM保护性自噬,抑制膀胱癌耐药细胞增殖,促进耐药细胞凋亡.%Objective: To investigate the effect of resveratrol on BIU-87/ADM autophagy in bladder cancer resistant cell line. Methods: Resveratrol was prepared at the concentration of 12.5μmol/L, 25.0μmol/L, 50.0μmol/L and 100μmol/L, respectively, and BIU-87/ADM cells of human bladder cancer were used as control. The effects of resveratrol at different concentrations on the proliferation of drug-resistant bladder cancer cell line BIU-87/ADM were detected by CCK-8 assay. The drug resistant cells of bladder cancer were detected by RT-PCR assay before and after treatment with resveratrol. The expression levels of LC-3 and Beclin-1 mRNA in BIU-87/ADM cells were detected by Wester blot. Results: The growth inhibition rate of BIU - 87/ADM cells was significantly higher than that in the control group at 24h and 48h. The higher the concentration of resveratrol was, the higher the apoptosis rate was. The expression of LC3-Ⅱand Beclin-1 mRNA decreased significantly after the intervention of resveratrol, and the higher the concentration of resveratrol, the more the degree of decrease of LC3-Ⅱ and Beclin - 1 mRNA expression. Conclusion: Resveratrol can significantly reduce the protective autophagy of drug resistant cell line BIU-87/ADM, inhibit the proliferation of drug resistant cell line of bladder cancer, and promote the apoptosis of drug resistant cell line.
