高效液相色谱法快速测定柑橘果皮中8种类黄酮

         

摘要

A high Performance Liquid Chromatography(HPLC) method was built for determination of 8 flavonoids in citrus peels (stcitrate glycosides, naringin, hesperidin, neohesperidin, didymin, narirutin, naringenin, hesperetin). Samples were extracted by methanol-acetone (70:30), then separated with methanol-0.2% acetic acid solution by gradient elution, finally detected by diode array detector. The results showed that,when the linear range was 0.5~200 mg/L, the concentrations of all compounds exhibited good linear relationship with peak areas, the correlation coefficient was from 0.9996 to 0.9998. The recoveries were 85.0%~95.7%, the relative standard deviation was 1.32%~4.83%, the method detection limit was 0.2~0.8 mg/L, the quantitative limit was 0.8~3.0 mg/L. This method is simple, low cost, good reproducibility and high accuracy and suitable for the determination of flavonoids in citrus peel.%建立同时测定柑橘果皮中8种类黄酮(圣枸橼苷、柚皮苷、橙皮苷、新橙皮苷、香风草甙、柚皮芸香甙、柚皮素、橙皮素)的高效液相色谱法.样品经甲醇-丙酮(70:30)混合溶液提取,以甲醇-0.2%乙酸水溶液为流动相,采用梯度洗脱,二极管阵列检测器检测.在线性范围为0.5~200 mg/L时,8种类黄酮的峰面积与浓度呈现良好的线性关系.相关系数为0.9996~0.9998,在三个不同的添加水平条件下,回收率为85.0%~95.7%,相对标准偏差为1.32%~4.83%,方法检出限为0.2~0.8 mg/L,方法定量限为0.8~3.0 mg/L.该方法操作简便,成本低,重现性好,准确度高,是测定柑橘果皮中类黄酮的有效方法.
