复方甘草酸苷对实验性溃疡性结肠炎大鼠白细胞介素4、白细胞介素17水平的影响

         

摘要

目的 通过检测三硝基苯磺酸/乙醇诱导的溃疡性结肠炎(UC)大鼠模型血清白细胞介素4(IL-4)、IL-17水平,探讨复方甘草酸苷对实验性UC大鼠的治疗作用.方法 成年SD大鼠40只,随机分成4组(n=10):正常对照组不做任何处理,其余3组用2.5%三硝基苯磺酸钠/乙醇溶液灌肠制备溃疡性结肠炎模型,模型组用生理盐水灌肠3天后处死,复方甘草酸苷治疗组每日用复方甘草酸苷溶液灌肠,40 mg·kg-1·d-1,连续灌肠7天后处死;柳氮磺胺吡啶(SASP)治疗组每日用SASP溶液灌肠,0.5 g· kg-1·d-1,连续灌肠7天后处死.经不同处理和治疗后,用酶联免疫吸附测定法检测各组大鼠血清中IL-4、IL-17的表达水平.结果 与正常组比较,模型组IL-4明显降低,IL-17明显升高,IL-4(81.52±8.21) ng/L vs (90.82±7.35) ng/L(P<0.01),IL-17(172.43±23.55) ng/L vs (135.26±27.86)ng/L(P<0.01);与模型组比较,复方甘草酸苷治疗组、SASP治疗组IL-4水平明显升高,IL-17水平明显降低,IL-4(88.45±8.58) ng/L、(87.53±7.65) ng/L vs (81.52±8.21) ng/L(P<0.01);IL-17(140.35±22.78) ng/L、(142.11±21.56) ng/L vs (172.43±23.55) ng/L(P<0.01).结论 复方甘草酸苷可升高实验性UC大鼠血清IL-4的表达水平,降低IL-17的表达水平,可能通过调控这两个细胞因子的表达而发挥疗效.%Objective To investigate the experimental therapeutic action of the compound glycyrrhizin on ulcerative colitis ( UC) in rats induced by trinitrobenzene sulfonic acid/ethanol ( TNBS/ethanol) through the examination of serum interleukin 4OL-4) and interleukin 17(IL-17) levels.Methods Forty adult Sprague-Dawley rats were randomly divided into four groups ( n =10), normal control group did not receive any treatment, the rest three groups were used trinitrobenzene sulfonic acid/ethanol enema to preparate ulcerative colitis model.Model group after using saline water enema for three days would be put to death,compound glycyrrhizin glycosides treatment group daily used compound glycyrrhizin glycosides solution enema, 40 mg·kg-1·d-1 , continuous enema 7 days put to death, SASP treatment group daily used SASP solution enema,0.5 g·kg-1·d-1 continuous enema 7 days put to death.Each group received different treatment as described above, the IL-17 and IL-4 in serum were observed through enzyme-linked immunosorbent assay(ELISA).Results Compared with those of normal control group, serum IL-4 in model group was significantly decreased,and serum IL-17 in model group was significantly increased, IL-4 (81.52±8.21) ng/L vs (90.82±7.35) ng/L( P <0.01), IL-17 (172.43±23.55) ng/L vs (135.26±27.86) ng/L( P <0.01); Compared with those of model group, serum level of IL-4 in compound glycyrrhizin glycosides treatment group was significantly increased,and serum level of IL-17 in compound glycyrrhizin glycosides treatment group was significantly increased, IL-4 (88.45±8.58) ng/L, (87.53±7.65) ng/L vs (81.52±8.21) ng/L(P <0.01);IL-17 (140.35±22.78) ng/L,(142.11±21.56) ng/L vs (172.43±23.55) ng/L( P<0.01).Conclusion Compound glycyrrhizin can increase serum level of IL-4 and decrease serum level of IL-17, its potential therapeutic effect may be modulating the expressions of these two cytokines.
