芳维A酸乙酯对银屑病皮损角质形成细胞凋亡和bax表达的影响

摘要

目的　探讨芳维A酸乙酯通过诱导细胞凋亡在银屑病治疗中发挥的作用。方法　用脱氧核苷转移酶介导的d-UTP-生物素缺口末端标记技术(TUNEL法)和免疫组化方法检测芳维A酸乙酯治疗前和治疗15d、治疗30d时银屑病皮损细胞凋亡和bax的表达。结果　芳维A酸乙酯治疗第15d和第30d，银屑病皮损细胞凋亡及bax表达均明显增加；图像定量分析和治疗前相比，均有显著性增加(P<0.01)；细胞凋亡的程度与bax的表达高度相关(r＝0.91)。结论　诱导细胞凋亡可能是芳维A酸乙酯治疗银屑病的重要机制之一，其作用与诱导凋亡基因bax的表达有关。%Objective　To investigate the involvement of apoptosis in psoriatic lesions with arotinoid ethyester treatment.Methods　The apoptosis was detected with terminal deoxynucleotidyl transferase-mediated d-UTP biotin nick end labeling(TUNEL), and the expression of bax was assessed with immunoperoxidase technique in keratinocytes of psoriatic lesions before and after treafment for 15 days and 30 days by arotinoid ethyester.Results　Arotinoid ethyester could induce keratinocytes of psoriatic lesions apoptosis quickly. Compared with the results of pretreatment, the degree and extent of apoptosis and expression of bax in keratinocytes of psoriatic lesions treated by arotinoid ethyester for 15 and 30 days were significantly increased(P<0.01),the degree of apoptosis was highly related to the expression of bax(r=0.91).Conclusion　The effect of apoptotic induction by arotinoid ethyester might play an important role in psoriatic treatment, it was related to the expression of apoptotic gene bax.
