Objective To study the expression of transforming growth factor-β2 (TGF-β2) and to investigate the epileptogenic mechanism and the mechanism of morphological changes in the hippocampus of the disorders of cortical developments (DCDs) rat models. Methods The DCDs rat models were established. The expression changes of TGF-β2 in the hippocampus of the P 60 d off-spring were analyzed by using immunohistochemistry, Western blot and quantitative PCR. Results Compared with the control group, the protein expression and the RNA expression of TGF-β2 in the hippocampus of the DCDs rat models were both significantly lower(the protein expression: 60 ± 0. 17 vs 0. 92 ± 0. 19;P<0. 05,the RNA expression: 48 ± 0. 07 vs 1. 12 ± 0. 16;P<0. 05). Conclusion These results suggest that the TGF-β2 can play an important role in disorder migration of hippocampal neurons in the DCDs rat models.%目的 检测转化生长因子-β2(TGF-β2)在皮质发育障碍模型大鼠海马的表达,探讨海马形态改变的发生机制及致癫机制.方法 制作皮质发育障碍大鼠模型,选取出生60 d幼鼠,利用免疫组化、荧光定量PCR和Western blot等方法检测TGF-β2在皮质发育障碍模型大鼠海马区的变化.结果 与正常对照组比较,皮质发育障碍组大鼠海马TGF-β2 mRNA和蛋白表达明显降低(蛋白:0.60±0.17 vs 0.92±0.19,P<0.05;mRNA:0.48±0.07 vs 1.12±0.16,P<0.05).结论 TGF-β2表达变化可能与皮质发育障碍大鼠海马神经元的迁移紊乱有关.
