Survivin基因沉默对裸鼠人卵巢癌细胞移植瘤凋亡的影响

             

摘要

目的 探讨survivin基因沉默对人卵巢癌细胞株SKOV3裸鼠移植瘤凋亡的影响.方法 将重组survivin shRNA plasmid转染人卵巢癌细胞株SKOV3细胞,通过裸鼠移植瘤实验比较survivin基因沉默对裸鼠移植瘤的生长抑制作用,荧光定量聚合酶链反应(PCR)和蛋白印迹法检测与凋亡密切相关的cytochrome-c、caspase-9和caspase-3基因的表达.结果 裸鼠移植瘤实验显示,与对照组(空载体组和未转染组)相比,survivin shRNA plasmid转染组人卵巢癌细胞株SKOV3细胞生长速度减慢,肿瘤体积缩小(P<0.05);荧光定量PCR和蛋白印迹法结果都显示,与对照组相比,survivin shRNA plasmid转染组cytochrome-c、caspase-9和caspase-3的表达明显升高(P<0.05).结论 survivin基因沉默可通过促进肿瘤细胞的凋亡而抑制裸鼠移植瘤生长.%Objective To investigate the effect of survivin gene silencing on tumor xenograft apoptosis of SKOV3, the human ovarian cancer cell line, in nude mice. Methods Human ovarian cancer cell line SKOV3 was transfected using recombinant survivin shRNA plasmid for further comparison of survivin gene silencing on tumor xenograft growth inhibition. Fluorescent quantitative polymerase chain reaction and Western blotting were applied to determine the expression of cytochrome-c, caspase-9 and caspase-3 that were closely related to apoptosis. Results Compared with control groups (unloaded-vector and untransfected group), the human ovarian cancer cell line SKOV3 transfected with survivin shRNA plasmid, as reflected by nude mice xenograft experiment, was found as having lower growth rate and tumor shrinkage (P<0.05). Fluorescent quantitative polymerase chain reaction and Western blotting showed that survivin shRNA plasmid transfection group yielded considerably up-regulated cytochrome-c,caspase-9 and caspase-3 expression than control group (all P<0.05). Conclusion Survivin gene silencing may inhibit growth of xenograft via induction of tumor cell apoptosis in nude mice.
