肝X受体在糖尿病大鼠肾损伤模型中的作用及意义

         

摘要

Objective To investigate the expression of liver X receptor (LXR) in the kidney at the different time points in streptozocin (STZ)-induced diabetic rats. Methods Forty male WISTAR rats were randomly assigned to control group and experimental group. The experimental group was intraperitoneally injected with STZ at a dose of 60 mg/kg. At weeks 4, 8 and 12, we determined the body weight, blood glucose, serum creatinine and 24-hour pro-teinuria. The level of LXR mRNA and protein expression at weeks 4, 8 and 12 was assayed by reverse transcriptase polymerase chain reaction and Western blotting technique. Results Results of reverse transcriptase polymerase chain reaction suggested that LXR expression was up-regulated at week 4 and culminated at weeks 12 (both P<0.05). West-ern blot further confirmed the above findings. Conclusion The down-regulated expression of LXR of renal tubular epithelial cells in experimental group has played a role in promoting renal injury in diabetic rats.%目的:观察肝X受体(LXR)在链脲佐菌素(STZ)诱导的不同时间段糖尿病大鼠肾脏中的表达情况。方法雄性Wistar大鼠40只,数字法随机分为对照组10只和实验组30只。实验组用链脲佐菌素(STZ)60 mg/kg一次性腹腔注射,分别于4、8、12周将实验组与对照组大鼠测定体质量、血糖、血肌酐、尿蛋白定量(24 h);实时荧光定量聚合酶链反应(RT-PCR)、蛋白印迹法测定4、8、12周LXR mRNA和蛋白的表达情况。结果RT-PCR结果提示,从实验第4周开始,LXR表达量开始提高,12周达到最高(P<0.05);蛋白印迹法结果可见4周开始增加,12周表达达到高峰。8周高于4周,12周高于8周(P<0.05)。结论实验组肾小管上皮细胞LXR的表达下调在糖尿病大鼠的肾损伤中起了促进的作用。
