白细胞介素-23受体基因多态性与强直性脊柱炎遗传易感性关系的荟萃分析

     

摘要

Objective To investigate the correlation between interleukin-23 receptor (IL23-R) gene polymor-phism and genetic susceptibility in ankylosing spondylitis (AS) by meta-analysis. Methods Internet search in databases including Cochrane, Pubmed, EMBASE, Web of Science, CNKI and CBM was performed to identify case-control studies on the correlation between IL-23R polymorphism and genetic susceptibility in AS for meta-analysis. Results A total of 11 studies involving 2652 AS cases, 3221 healthy controls, and 4 study populations were included in the meta-analysis. The meta-analysis showed: ①Rs10489629 gene polymorphism was significantly correlated with AS genetic susceptibility in general study subjects (OR=0.84, 95%CI 0.74-0.96, P=0.009). It was also significantly correlated with AS genetic susceptibility in Mongolians based on ethnicity stratification (OR=0.76, 95%CI 0.60-0.97, P=0.02). ②Rs1343151 and rs11209026 gene polymorphisms were significantly correlated with AS genetic susceptibil-ity in general study subjects (OR=0.68, 95%CI 0.58-0.80, P<0.001; OR=1.99, 95%CI 1.41-2.81, P<0.001). They were correlated with AS genetic susceptibility Caucasians based on ethnicity stratification, respectively ( OR=0.67, 95%CI 0.56-0.79, P<0.001; OR=2.23, 95%CI 1.49-3.35, P<0.001). ③No correlations were found between rs1004819, rs11209032, rs10889677, rs7517847, rs1495965 and AS genetic susceptibility. Conclusion IL-23R gene poly-morphism may be correlated with genetic susceptibility in AS. The related polymorphic sites are rs10489629, rs1343151 and rs11209026. However, significant differences are found in the correlation of IL-23R gene polymor-phism and AS genetic susceptibility between different ethnic groups.%目的:用荟萃分析方法了解白细胞介素-23受体(IL-23R)基因多态性与强直性脊柱炎(AS)遗传易感性之间的相关性。方法通过检索Cochrane图书馆、Pubmed数据库、EMBASE数据库、Web of Science数据库、中国期刊全文数据库(CNKI)和中国生物医学文献数据库(CBM)搜集IL-23R基因多态性与AS遗传易感性相关的病例对照研究进行荟萃分析。结果11篇文献,累计2652例AS病例、3221名健康对照,4个研究人群纳入本次荟萃分析。荟萃分析结果提示:①rs10489629的基因多态性在总体人群中与AS遗传易感性存在相关性[OR=0.84,95%CI(0.74,0.96),P=0.009];按人种分层则与蒙古人种AS遗传易感性存在相关性[OR=0.76,95%CI(0.60,0.97),P=0.02]。②rs1343151和rs11209026的基因多态性在总体人群中显示出与AS遗传易感性的相关性[OR=0.68,95%CI(0.58,0.80),P<0.01];[OR=1.99,95%CI(1.41,2.81),P<0.01];按人种分层则与高加索人种的AS遗传易感性存在相关性[OR=0.67,95%CI(0.56,0.79),P<0.01];[OR=2.23,95%CI(1.49,3.35),P<0.01]。③rs1004819、rs11209032、rs10889677、rs7517847和rs1495965与AS遗传易感性不存在相关性。结论 IL-23R基因多态性可能与AS遗传易感因素相关,相关的多态性位点包括rs1343151、rs10489629和rs11209026。但IL-23R基因多态性与AS遗传易感性的关联存在明显的人种差异。
