丝素/辛伐他汀复合膜修复大鼠颅骨缺损的实验研究

         

摘要

Objective To evaluate the effect of silk fibroin membrane, a scaffold material, loaded simvastatin on osteogenesis in repairing the rat calvarial defects, and to investigate its feasibility and effect on the repair of bone defects. Methods The prepared silk fibroin/simvastatin membrane was placed on the bilateral 5 mm bone defects of the rat skull. At 4 and 8 weeks after the implantation, the gross observation, HE staining and immunohistochemical staining were used to determine the bone morphological changes and the expression of BMP-2 and ALP, and the bone healing was evaluated. Its repair effect was compared with that in the simple silk fibroin membrane implantaion group. Results Compared with the simple silk fibroin membrane implantaion group, the osteoblasts grew more active in the scaffolds, and the osteogenesis of the defect area was more obvious, and there were more positive expression of BMP-2 and ALP in the defect area in the silk fibroin/simvastatin composite membrane group. There were statitically signifi-cant differences between the two groups. Conclusion Silk fibroin/simvastatin composite membrane can effectively promote osteogenesis at bone defect and promote bone healing by up-regulating the expression of BMP-2 and ALP, which has potential application value in promoting bone healing.%目的 旨在评估丝素膜作为一种支架材料并加载辛伐他汀对修复大鼠颅骨缺损区成骨的影响并探讨其对骨组织缺损修复的可行性及效果.方法 将制备好的丝素/辛伐他汀复合膜置于大鼠颅骨双侧5 mm骨缺损处,在植入后4、8周时,应用大体观察、苏木精-伊红(HE)染色及免疫组织化学染色的方法,观察骨缺损区的组织学形态变化及骨形成蛋白(BMP)-2和碱性磷酸酶(ALP)的表达情况来评价其骨再生.并与单纯丝素膜植入组的修复效果进行比较.结果 丝素/辛伐他汀复合膜植入组较单纯丝素膜植入组相比,支架内成骨细胞生长更活跃,缺损区边缘成骨更明显;缺损区BMP-2和ALP的阳性表达更多,差异有统计学意义.结论 丝素/辛伐他汀复合膜能有效促进骨缺损处成骨并通过上调BMP-2和ALP的表达来促进骨再生,因此对促进骨愈合具有潜在的应用价值.
