肝脏G蛋白偶联受体与糖代谢调节

         

摘要

肝脏是调节机体能量平衡尤其是糖、脂代谢的主要器官.在前糖尿病状态,患者表现为高胰岛素血症以及包括肝脏在内的外周组织的胰岛素抵抗,但不表现为空腹血糖升高.随着病程的发展,患者自身代偿能力逐渐衰退,出现肝脏糖代谢紊乱,糖原分解和糖异生增强,空腹血糖水平增高,最终发生糖尿病.研究表明,一些G蛋白偶联受体(GPCRs)参与了肝脏糖异生、糖原分解等代谢途径的调节,在肝糖代谢中发挥着重要作用.该文综述了参与肝糖代谢调节作用的GPCRs,以及靶向GPCRs改善胰岛素抵抗、治疗2型糖尿病的研究进展.%Liver, a key organ in energy balance, plays an important role in regulating glucose and lipid metabolism.In pre-diabetic status, patients show hyperinsulinemia and insulin resistance in peripheral tissue ,including the liver, but do not show for the fasting hyperglycemia.As the disease progresses, the liver glucose metabolism disorder appears, excessive glycogenolysis and gluconeogenesis arise, Which results in hyperglycemia.Several G protein-coupled receptors ( GPCRs ) expressed in liver tissues are known to be involved in the regulation of glycogenolysis and gluconeogenesis.Here,the progresses of the GPCRs involved in regulation in glucose metabolism and the targeting GPCRs for insuline resistance and the diabetes have been reviewed.
