高效液相色谱与串联质谱法测定游离丙戊酸钠与人血浆蛋白的结合率

摘要

目的：建立高效液相色谱与串联质谱法，考察丙戊酸钠在人血浆中的蛋白结合率。方法：采用中空纤维前处理技术结合高效液相与串联质谱法，测定游离丙戊酸钠的浓度，采用蛋白沉淀法测定人血浆中总的丙戊酸钠的浓度。结果：低、中、高3种浓度下，丙戊酸钠的血浆蛋白结合率分别为88.27%、89.63%、95.55%。结论：建立的方法经方法学考察能满足药品分析要求。%Objective: To establish a high performance liquid chromatography (HPLC) with tandem mass spectrometry to study the binding rate of sodium valproate to plasma proteins. Methods: The hollow fiber for pretreatment combined with HPLC-MS/MS was used to determine free sodium valproate. The protein precipitation method was used for determination of total sodium valproate in human plasma. Results:The plasma protein binding rates of sodium valproate at low, middle and high concentrations were 88.27%, 89.63% and 95.55%. Conclusion:In accordance with the requirements of the methodology veriifcation, the method can be used for drug analysis.
