Objective To investigate the expression and clinical significance of matrix metalloproteinases-9 ( MMP-9 ) and tissue inhibitor of metalloproteinases-1 ( TIMP-1 ) in placenta of patients with severe preeclampsia. Methods Sixty pregnant women hospitalized in department of gynecology and obstetrics of Xijing Hospital from August 2010 to February 2011 were enrolled in the study, and they were divided into two groups: severe preeclampsia group ( 30 cases ) and control group ( 30 cases ). The expressions of MMP-9 and TIMP-1 in placenta of severe preeclampsia patients and normal controls were detected with immunohistochemistry and western blot. Results The level of MMP-9 in severe preeclampsia group was significantly lower than that in the control group ( 0.04 ±0.01 vs 0. 12 ±0. 03 , t = 5. 70, P < 0.01), but the level of TIMP-1 was significantly higher than that in control group ( 0. 10 ± 0. 01 vs 0. 02 ±0. 003 , t = 4. 42, P < 0.01 ). Meanwhile, the ratio of MMP-9 /TIMP-1 was lower in severe preeclampsia group than in control group ( 0.40 ±0. 12 vs 8. 11 ± 2. 88 ,t = 3.30,P<0. 01 ). Conclusion The abnormal expression of MMP-9 and TIMP-1 in the placenta of severe preeclampsia patients might be related to insufficient trophoblast infiltration.%目的 研究基质金属蛋白酶-9(matrix metalloproteinases-9,MMP-9)和金属蛋白酶组织抑制因子-1(tissue inhibitor of metalloproteinases-1,TIMP-1)在重度子痫前期患者胎盘中的表达及临床意义.方法 选择2010年8月至2011年2月在西京医院妇产科住院分娩的孕妇60例,其中重度子痫前期组30例、正常妊娠(对照)组30例,采用免疫组化法、蛋白印迹法检测重度子痫前期患者和正常妊娠妇女胎盘滋养细胞中MMP-9、TIMP-1的表达.结果 在重度子痫前期组胎盘滋养细胞中,MMP-9的表达比正常胎盘滋养细胞中显著下降(0.04±0.01vs 0.12±0.03,t=5.70,P<0.01),TIMP-1的表达则比正常胎盘滋养细胞中显著升高(0.10±0.01vs 0.02±0.003,t=4.42,P<0.01),且MMP-9 /TIMP-1的比值明显低于在正常妊娠胎盘中的表达(0.40±0.12 vs 8.11±2.88,t=3.30,P<0.01).结论重度子痫前期患者胎盘组织中MMP-9、TIMP-1的异常表达可能与滋养细胞侵入障碍相关.
