香蕉胚性细胞悬浮培养方法的改进

     

摘要

针对香蕉胚性悬浮细胞培养过程中存在的胚性愈伤组织的诱导率低、周期过长、胚性易丢失等问题,以“苹果蕉”及“威廉斯”香蕉的未成熟雄花为试验材料,对香蕉胚性愈伤组织的诱导、悬浮培养的理化条件进行改进.结果表明:(1)光照条件较暗培养更有利于诱导理想的香蕉胚性愈伤组织;(2)M1培养基中加入酪蛋白和脯氨酸有利于胚性愈伤组织的诱导;(3)用微孔板代替三角瓶悬浮培养,有利于悬浮细胞分裂增殖,而且可减少污染的可能.%Development of banana embryogenic cell suspension cultures is constrained by factors such as low induction frequencies of embryogenic callus, long induction period, and loss of embryogenic competence. In this paper, we reported on the improvement of induction conditions of embryogenic callus for the establishment of an efficient and reliable method of banana embryogenic cell suspension, using immature male flowers of "Musa spp. AAB cv". "Dwarf Brazilian " and " Musa spp. AAA cv" "Williams" as initial explants. Three conclusion points could be drawn from the present work: (1 Might conditions was good for banana embryogenic callus inducing; (2) adding Casein and Proline in the M, medium could help to improve the induction rate of embryogenic callus; (3) using microwell plates instead of Erlenmeyer flasks to initiate cell suspension cultures not only enhanced cell division and proliferation, but also reduced the contamination potentials.
