乌头注射液关节腔注射对膝骨关节炎模型兔关节液IL-1β、IL-18、NF-κB p65的影响

         

摘要

目的:观察关节腔注射乌头注射液对膝骨关节炎模型兔关节液IL-1β、IL-18、NF-κB p65等指标的影响,初步探讨乌头注射液治疗骨性关节炎的可能作用机理.方法:50只兔随机分为正常组、模型组、玻璃酸钠组、正清风痛宁组和乌头注射液组,除正常组外其余各组兔经关节腔注射木瓜蛋白酶诱导骨关节炎模型,2周后各组分别于第1、4、7、10d共4次关节腔注射给药,1周后处死兔子取膝关节软骨组织,光镜下观察组织形态学改变并进行Mankin评分,采用Elisa法检测关节液IL-1β、IL-18、NF-κBp65含量.结果:鸟头注射液组关节软骨病理改变Mankin评分总积分较模型组和玻璃酸钠组明显降低,乌头注射液组关节液中IL-1β、IL-18、NF-κB p65含量较模型组和玻璃酸钠组均明显降低,IL-18含量较正清风痛宁组明显减低,组间比较均有统计学意义(P<0.05).结论:乌头注射液对膝关节骨性关节炎模型兔的关节软骨病理改变有一定作用,可能与其抑制关节液中炎症因子IL-1β、IL-18、NF-κBp65的表达有关,其作用可能与激活NF-κB信号通路有关.%Objective:To observe the effect of Intra-articular injections of Wutou Injection on IL-1β,IL -18 and NF-B p65 in synovial fluid of rabbit model of knee osteoarthritis,and to explore the possible mechanism of Wutou injection in the treatment of osteoarthritis.Methods:50 rabbits were randomly divided into normal group,model group,Sodium Hyaluronate group,Zhengqing Fengtongning group and Wutou Injection group.Except the normal group,the other groups were injected with papain induced osteoarthritis modeL After 2 weeks,the rabbits in each group were injected with drug for4 times on the 1st,4th,7th,10th day.1 week later,rabbits were killed and cartilage tissue of knee joint was taken.The morphological changes and Mankin score were observed under light microscope.The contents of IL-1β,IL-18,NF-κB p65 in synovial fluid were detected by Elisa.Results:The total score of Mankin score in articular cartilage was significantly lower than that in model group and sodium hyaluronate group,and the contents of IL-1β,IL-18 and NF-κB p65 in synovial fluid of Wutou Injection group were significantly lower than those in model group and the sodium hyaluronate group,the contents of IL-18 in synovial fluid of Wutou Injection group were significantly lower than that in Zhengqing Fengtongning group.The differences between groups were statistically significant (P < 0.05).Conclusion:Wutou Injection may play an important role in the pathological changes of articular cartilage in rabbits with knee osteoarthritis model.It may be related to its inhibition of IL-1β,IL-18 and NF-κB p65 expression in synovial fluid,and its effect may be related to activation of NF-κB signaling pathway.
