创伤修复过程中创缘皮肤细胞因子含量变化与损伤时间的关系

     

摘要

BACKGROUND: The role of cytokines in wound healing process has been recognized in recent years, but it is not so clear about the expression changes of these cytokines in wound healing process as well as their relationship to wound age.OBJECTIVE: To explore the relationship between the expression change of cytokines and the wound age during the healing process of rats skin wound.DESIGN: A completely randomized controlled trial.SETTING and PARTICIPANTS: Twenty wistar rats, weighted 175 - 200 g,purchased from the Experimental Animal Center of the First Military Medical University.INTERVENTIONS: Animal models were prepared by making incisions bilateral to the vertebral column and the level of cytokines were measured with enzyme-linked immunosorbent assay.MAIN OUTCOME MEARSURES: The contents of interleukin-2(IL-2),tumor necrosis factor alpha(TNF-α) in skin samples of experimental and control groups.RESULTS: The level of IL-2 and TNF-α increased at 0. 5 hour after wounding and a peak of IL-2 and TNF-α occurred at 3 hour and 1 hour after injury respectively. Rebound of TNF and IL-2 levels were shown at 48 hour after wounding, and both cytokine levels were inclined to elevating between 72 hours and 168 hours after wounding.CONCLUSION: The cytokines have a time-related expression during wounds healing process.%背景:细胞因子在创伤修复中的作用日益受到重视,但诸多细胞因子在创伤修复中的表达变化及其与损伤时间的关系还有待进一步研究.目的:了解细胞因子在创伤及修复过程中的表达变化与损伤时间的关系.设计:完全随机实验对照研究.地点和对象:实验地点在华中科技大学同济医学院法医系完成,对象为清洁级Wistar大鼠20只,体质量175~200 g,雌雄不拘,由解放军第一军医大学提供.干预:实验动物背部造创制作动物皮肤创伤模型,用免疫酶联吸附方法(ELISA)检测细胞因子表达水平.主要观察指标:各组创缘皮肤白细胞介素2(IL-2)、肿瘤坏死因子(TNF-α)含量.结果:IL-2,TNF-α在造创后0.5 h即升高,并分别在造创后3 h和1 h到达峰值,在造创后48 h均有反弹,在72和168 h持续升高.结论:上述细胞因子在创口修复中的蛋白水平表达呈时间相关性特点.
