GPC5基因表达水平与肺腺癌淋巴结转移

摘要

目的 探讨磷脂酰肌醇蛋白聚糖-5(GPC5)在非小细胞肺腺癌中的表达水平及其临床意义.方法 应用Real-time PCR的方法检测134例非小细胞肺腺癌组织及其对应的正常肺组织中的GPC5基因的表达情况.应用免疫组化方法验证GPC5蛋白在人肺腺癌组织芯片(包含75对腺癌组织标本)中的表达情况.结果 (1) GPC5 mRNA表达水平在非小细胞肺腺癌组织中较正常肺组织显著下降(P＜0.001).(2)有淋巴结转移组较无淋巴结转移组GPC5 mRNA和蛋白表达水平均显著降低(P＜0.05).(3) Spearman相关性分析显示GPC5 mRNA表达水平与淋巴结转移、病理分期和分化程度呈显著正相关(P＜0.05).(4) Kaplan-Meier生存曲线分析示GPC5蛋白表达与肺腺癌患者长期预后相关(P =0.036).结论 GPC5的表达水平与非小细胞肺腺癌的淋巴结转移密切相关,可作为评估预后的辅助标志物.%Objective To investigate glypican-5 (GPC5) expression in Non-small cell lung adenocarcinoma and its clinical significance.Methods GPC5 was detected in 134 cases of Non-small cell lung adenocarcinoma and corresponding normal lung tissue by Real-time PCR,which was also validated in human lung adenocarcinoma tissue microarrays (TMA,containing 75 cases of lung adenocarcinoma tissue and adjacent tissue) by immunohistochemistry.Results (1) GPC5 mRNA expression level in Non-small cell lung adenocarcinoma was significantly lower than in matched normal lung tissue (P ＜ 0.001).(2)The expression levels of GPC5 mRNA and proteins in the lymph node metastasis group were all lower than those in the non-metastasis group(P ＜0.05).(3) Spearman analysis showed that the expression levels of GPC5 mRNA was significantly correlated with lymph node metastasis,TNM staging and differentiation.(4) The Kaplan-Meier survival analysis showed that the expressions of GPC5 proteins were related to the long-term prognosis of Non-small cell lung adenocarcinoma patients (P =0.036).Conclusion The expression levels of GPC5 were closely related with the lymph node metastasis of Non-small cell lung adenocarcinoma.It may be used as a marker of lung cancer diagnosis.
