母爱剥夺的成年大鼠伏隔核DNA甲基转移酶的表达

摘要

Objective To explore the effect of maternal deprivation on the expression of dopamine receptor (DRD2) in adult rats' nucleus accumbens and whether DNA methylation was involved in its expression regulatory mechanism.Methods Sixteen newborn rats were randomly divided into two groups,the maternal deprivation group ( n = 8 ) were deprived from their mother 6 hours per day during lactation,while the controls (n =8) without any treatment.When the rats grew up to twelve weeks, DRD2, DNAmethyltransferase mRNA expression level in nucleus accumbens was detected with real-time RTPCR.Results The expression level of DRD2 mRNA in maternal deprivation rats (1.25 ±0.06 ) was significantly lower than that in controls (1.15 ± 0.07) (P<0.05).However, the DNA methylransferase 1[(1.17±0.10) vs.(1.17±0.14)] and DNAmethyltransferasc3a [(1.18±0.07) vs.(1.20±0.16),P > 0.05] were not significantly different between both the groups.Conclusion DNA methylation may not be involved in the expression of gene DRD2.%目的 探讨DNA甲基化是否参与调控伏隔核多巴胺受体2(DRD2)基因表达,探索DNA甲基转移酶(Dnmts)是否参与调控DRD2基因的表达.方法 采用随机数字表法对16只新生大鼠随机分为母爱剥夺组和对照组,每组8只.母爱剥夺组大鼠于授乳期每天接受6 h母爱剥夺,12周龄时采用实时定量聚合酶链反应检测伏隔核DRD2受体、DNA甲基转移酶mRNA表达水平.结果 母爱剥夺组大鼠脑内伏隔核DRD2 mRNA表达水平(1.25±0.06)低于对照组(1.15±0.07;t=2.83,P<0.05);母爱剥夺组大鼠和对照组大鼠脑内伏隔核DNA甲基转移酶1 mRNA表达水平分别为1.17±0.10和1.17±0.14(t=-0.14,P>0.05);DNA甲基转移酶3a mRNA表达水平分别为1.18±0.07和1.20±0.16(t=0.39,P>0.05);DNA甲基转移酶3b无法检测.结论 DNA甲基化可能不参与调控伏隔核DRD2基因的表达.
