鞘脂激活蛋白原及其受体GPR37在口腔鳞癌中的组织学表达研究

         

摘要

目的 研究鞘脂激活蛋白原(prosaposin,PSAP)及其受体G蛋白偶联受体(G-protein coupled receptor,GPR)37在口腔鳞癌组织中表达及其与临床因素之间的关系和意义.方法 采用免疫组化技术检测41例口腔鳞癌标本中PSAP及GPR37的表达情况.通过使用CD68和PSAP或GPR37抗体及免疫荧光技术检测PSAP及GPR37的具体定位.结合临床资料,对照临床病理特征进行统计分析.结果 PSAP及其受体GPR37在口腔鳞癌中的表达明显高于周围正常组织(P<0.05),且PSAP的表达与肿瘤TNM分期以及颈部淋巴结转移之间有明显关联,而GPR37的表达则与上述临床指标无关.免疫荧光结果显示,PSAP在口腔鳞癌组织CD68阳性细胞(即单核巨噬细胞)中呈高水平表达,而GPR37在单核巨噬细胞表达不明显.结论 PSAP及其受体GPR37在口腔鳞癌中表达增强,可能与口腔鳞癌的发生发展有关.%Objective To study the expression of prosaposin (PSAP)and its receptor G-protein coupled receptor (GPR)37 in oral squamous cell carcinoma (OSCC),and its relationship with clinical characteristics of OSCC.Methods The expressions of PSAP and GPR37 were observed in 41 samples of oral squamous cell carcinoma by immunohistochemistry.The location of PSAP and GPR37 was determined by immunofluorescence with the antibodies of CD68,PSAP and GPR37.Based on the clinical data,the clinical pathological features were statistically analyzed.Results The expression of PSAP and GPR37 was higher in the OSCC than in normal oral mucosa(P < 0.05),and the expression of PSAP was closely related to tumor stage and lymph-node metastasis,but GPR37 was not.PSAP rather than GPR37 was strongly expressed in the macrophage cells,as the immunofluorescence results showed.Conclusion The expression of PSAP and its receptor GPR37 increases in the OSCC,which may be associated with the development of squamous cell carcinoma.
