间歇性张应力对小鼠单核/巨噬细胞RAW264.7分化成熟的影响

     

摘要

Objective: To study the effect of the differentiation and maturation of murine monocytes/macrophage of RAW 264.7 cells by the tensile stress. Methods:Pressing intermittent tensile stress with 1000με, 3000με, 5000με to the induced differentiation in vitro of RAW 264.7 cells, using tartrate-resistant acid phosphatase(TRAP) staining and acridine orange staining to observe the osteoclast forma-tion and analyse osteoclast differentiation status. Result: The tensile stress promotes the proliferation and differentiation of osteoclasts and the TRAP positive cells increased, with increasing the tensile stress the number of TRAP positive cells showed an increasing trend, but when tensile stress increased to 5000 μεTRAP positive cells did not increase obviously. Conclusions: The intermittent tensile stress induced murine monocytes/macrophage of RAW 264.7 cells into osteoclasts.%目的:探讨张应力对破骨前体细胞小鼠单核/巨噬细胞株RAW264.7分化成熟的影响。方法:施加1000με、3000με、5000με间歇性张应力于体外诱导分化的RAW264.7细胞,采用抗酒石酸酸性磷酸酶(TRAP)染色和吖啶橙染色观察破骨细胞形成情况,并分析比较各组破骨细胞分化状况。结果:牵张应力促进了破骨细胞的增殖和分化,其TRAP阳性细胞增多,且随着牵张力量的增强TRAP阳性细胞数量呈增加趋势,但张应力增加到5000μεTRAP阳性细胞数量增加不明显。结论:间歇性的牵张力促进了小鼠单核/巨噬细胞RAW264.7向破骨细胞的分化。
