质谱技术分析联合用药抗大鼠肝纤维化的作用

         

摘要

目的:应用表面增强激光解析电离飞行时间质谱(surface enhanced laser desorption/ionization time of flight mass spectrometry,SELDI-TOF-MS)技术分析联合用药抗大鼠肝纤维化的肝组织中蛋白质的表达差异.方法:采用CCl4造成大鼠肝纤维化模型,试验分为正常对照组、模型组和联合用药治疗组.在试验的第10周末取大鼠肝组织,提取肝组织总蛋白质,应用SELDI-TOF-MS检测肝组织差异表达的蛋白.结果:SELDI-TOF-MS共检测到176个蛋白峰,其中表达差异2倍以上的蛋白有12个,包括在模型组中表达有降低趋势的10个蛋白质和升高趋势的2个蛋白质;经过联合用药治疗后,这些蛋白表达恢复到正常水平.对这12个差异表达蛋白进行蛋白质组数据库的检索,获得4个蛋白质,涉及负调控细胞生长、抑制细胞增殖和参与细胞信号转导.结论:联合用药能够通过调节大鼠肝脏组织蛋白质的表达,改善和修复肝脏功能.%Objective; To detect protein expression differences in liver tissue of rats with liver fibrosis after combination therapy by surface-enhanced laser desorption ionization time of flight mass spectrometry ( SELDI-TOF-MS ) . Methods: Liver fibrosis in rats was induced with CC14 treatment. Three groups of rats, named as normal control, liver fibrosis model and combination therapy groups, were used in this experiment. After 10 weeks, the rats were sacrificed and total protein was extracted from the liver tissue for the SELDI-TOF-MS detection. Results;A total of 176 protein peaks were detected and 12 of them had more than two times difference in expression levels, a-mong which 10 had decreased expression and 2 had enhanced expression in model group. After treatment with combination therapy,the expression turned normal. Among the 12 proteins, 4 were retrieved in the proteome database, which were related to the negative regulation of cell growth inhibition of cell proliferation, and signal transduction. Conclusion; Combination therapy can repair and improve liver function in rats through regulating the expression of liver protein.
