腹腔内注射L-精氨酸诱导法建立急性胰腺炎小鼠模型的评价

         

摘要

Objective  To investigate the method of making AP mice model induced by intraperitoneal injection of L-Arg .Methods  100 healthy male Kunming mice were divided randomly into normal contrast group and model group ,there were 10 mice respec-tively at 6h ,12h ,24h ,48h ,72h after L-Arg injection in every group .After intraperitoneal injection of L-Arg(200 mg/100g body weight) twice at an intervalofone hour ,the serum amylase was measured and the pancreatic inflammatory pathologic reaction was examined and scored at different time .AP mice model was proved successful by the above method .Results  After intraperitoneal injection of L-Arg ,the serum amylase level was gradually going up ,began to rise at 6h ,raised to peak at 12h ,got right at 72h .At different time the serum amylase level of model group was compared with normal contrast group and the difference existed notable significance(P < 0 .01) .By naked eye observing and the pathologic scoring analysis ,at 48 h the pancreas inflammatory changes were the most severe and the pancreatitis histological score was highest ,at 72h the histological scoring value was reduced .Conclusion  The AP mice model induced by intraperitoneal injection of L-Arg(200 mg/100 g body weight) twice at an interval of one hour ,accords with human pathologic process of AP basically ,and is one good model to research the AP pathogenesy .%目的 研究腹腔内注射L-精氨酸(L-arginine,L-Arg)诱导急性胰腺炎(acute pancreatitis,AP)小鼠模型的造模方法.方法 选择健康雄性昆明小鼠100只,分为正常对照组与AP模型组,每组在6 h、12 h、24 h、48 h、72 h各时间点各分10只.间隔1h两次于小鼠腹腔内注射L-精氨酸200 mg/100 g体重,在各时间点检测血清淀粉酶、进行病理检查并评分分析,证实动物模型成功.结果 小鼠腹腔内注射L-Arg后,血清淀粉酶呈逐渐上升的趋势,6 h血清淀粉酶活性开始升高,12 h达到高峰,72 h恢复正常.在各时间点造模组小鼠血清淀粉酶分别与正常组比较,差异有显著性(P<0.01).肉眼观察和病理评分分析结果显示,注射L-Arg后48 h胰腺炎症病变最为严重,病理评分分值最高,72 h胰腺组织病变程度降低.结论 间隔1 h两次于小鼠腹腔内注射L-Arg 200 mg/100 g体重制备的AP模型,基本符合人类AP病理过程,是研究AP发病机制良好的动物模型.
