柯萨奇B4病毒5′UTR基因序列及遗传进化分析

     

摘要

目的 研究柯萨奇B4病毒(CVB4jlu06株)5′UTR基因序列及变异性,探讨其致病性的分子生物学基础.方法 Hep-2细胞扩增病毒,RT-PCR扩增5′UTR基因片段,测序后进行同源性、变异性及遗传进化分析.结果 CVB4jlu06株5′UTR基因全长743个核苷酸,其与Tuscany株和JVB株有同源性较高,遗传距离较近;CVB4jlu06株5′UTR基因与CVB4致糖尿病株(E2株、E2b株和Tuscany株)有三个共同核苷酸.结论 CVB4jlu06株5′UTR基因序列与CVB4JVB株和CVB4Tuscany亲缘关系密切;CVB4jlu06株5′UTR 发生的核苷酸变异可能与其致病性相关.%Objective To study the 5 UTR nucleotide sequence and variation of Coxsackievirus B4 jlu06 strain,and characterize the genetic determinants involved in pathogenic phenotype. Methods Coxsackievirus B4 was amplified with Hep 2 cell. The 5 UTR gene fragment was amplified by RT PCR and sequenced,the mutations, homology and phylo genesis were analyzed. Results The 5 UTR of thejlu06 strain is 743 nucleotides (nt) long,which showed high homolo gies and short genetic distance to Tuscany strain and JVB strain. The 5 UTR nucleotide sequence of CVB4 jlu06 strain showed common amino acids with diabetogenic strains(E2 ,E2b and Tuscany strains) at 3 sites. Conclusion The 5 UTR of the jlu06 strain is closely related to CVB4Tuscany and CVB4JVB strain; The mutations in 5 UTR nucleotide sequence might be responsible for the pathogenic phenotype of this strain.
