结核分枝杆菌生物被膜体外模型的建立

摘要

Objective This study to explore how to establish model of tuberculosis bacterium biofilm in vitro.Methods Select the non-drug-resistant TB bacteria and resuscitate them in Roche solid cultur e medium.Then culture then for one week in the 7H9 OADC medium in order to reach the same activity.Culture the TB bacteria in screw capped bottle and orifice pl ate with Sauton's media.Results In orifice plate,we successfull y obtained biolfilm for all bacterial when used sealing film.In screw capped bot tle,we need to tight the cap until the end of the third week and loose the cap a t the end of the third week.We can observed the biofilm at the end of the second week and found the structure matured by the end of the fifth week.Concl usion TB biofilm can be successfully cultured in vitro.%目的 本试验探讨如何建立结核菌生物被膜体外模型.方法 选取生长良好的非耐药结核菌在罗氏固体培养基中复苏,在7H9 OADC培养基中重新培养1周,使其活力相同,配置苏通培养基分别在烧瓶及孔板中培养结核菌观察生物膜的生长情况.结果 36株结核菌在孔板中培养时使用封口膜的全部培养出生物膜,而未使用封口膜的均培养失败.烧瓶中培养:在培养3周时需要松开瓶盖,否则生物膜生长不良.时间:培养2周时可看到有生物膜出现,5周时生物膜培养成熟.结论 结核菌有生物被膜,通过体外培养可以成功获得.
