骨髓间充质干细胞与血管内皮生长因子联合移植对大鼠创伤性脑损伤修复的研究

         

摘要

目的 探讨联合移植骨髓间充质干细胞(BMSCs)和血管内皮生长因子(VEGF)对创伤性脑损伤(TBI)大鼠脑组织中脑源性神经营养因子(BDNF)表达的影响.方法 SD大鼠2只,取股骨骨髓体外培养BMSCs.SD大鼠40只,随机分为单独移植BMSCs组、单独应用VEGF组、联合移植BMSCs与VEGF组及对照组,每组10只.采用改良的Feeny自由落体撞击方法建造中度创伤性脑损伤模型.造模后1周,在立体定向仪下分别将等量BMSCs、VEGF、BMSCs加VEGF及PBS液注入相应大鼠的损伤脑组织周边.养育2周后处死取脑.采用HE及尼氏染色法观察脑组织病理学变化;采用免疫组化染色,应用Image pro-Plus6.0软件测量半暗带及健侧对称部位BDNF阳性表达区域的总和累积光密度,应用SPSS 11.3软件进行统计学分析.结果 除BMSCs组外,其余各组对称部位BDNF含量均较半暗带高(P<0.05);联合移植组半暗带的BDNF含量明显高于对照组(P<0.05).结论 联合移植BMSCs与VEGF可有效抑制TBI大鼠半暗带中BDNF含量的降低,有利于神经功能的恢复.%Objective To investigate the influence of transplantation of bone marrow mesenchymal stem cells (BMSCs) and vascular endothelial growth factor (VEGF) on the brain derived neurotrophic factor (BDNF) expression in rat brain with traumatic brain injury (TBI). Methods Femoral bone marrows for culturing BMSCs in vitro were obtained from 2 Sprague - Dawlcy (SD) rats. Forty SD rats were randomly divided into 4 groups: Individually transplanting BMSCs group,individually transplanting VEGF group, transplanting BMSCs and VEGF group and the control group, 10 rats in each group. Moderate traumatic brain injury model was constructed by using the improved Fecny free-falling impact method. After modeling for 1 week, the same amount of BMSCs, VEGF, BMSCs plus VEGF and PBS fluid were injected into the corresponding rat brain tissue surrounding the injury in stcrcotactic instrument. Two weeks later,the rats were put to death and the brain tissues were obtained. The pathological changes of brain tissue were observed by Hematoxylin - cosin (HE) and Nissl staining. With immunohistochemical staining, the sum cumulative optical density of the BDNF positive expression regions in the penumbra and the uninvolvcd symmetric parts were measure using Image pro - Plus6. 0 software. SPSS 11.3 software was used for statistical analysis. Results The BDNF contents in symmetrical parts were higher than the contents of penumbras except the BMSCs group(P<0. 05). The BDNF contents in the penumbras in transplantation group was significantly higher than that in control group(P<0. 05). Conclusion Combined transplantation of BMSCs and VEGF could effectively inhibit the decrease of BDNF contents of TBI rat penumbras. It was beneficial to the recovery of neurological function.
