去窦弓神经对大鼠神经细胞线粒体Ca2+和膜电位的影响

         

摘要

Objective To explore the effects of sinoaortic denervation (SAD)on mitochondrial Ca2+ and membrane potential of nerve cel in rats. Methods Fifty Sprague Dawley (SD)rats were randomly divided into control group,sham operation group,30 day SAD group,60 day SAD group and 90 day SAD group. Mitochondrial membrane potential (by Laser scanning confocal micro-scope)and level of Ca2+(by flame atomic absorption spectrometry)were measured and compared between the five groups.Results There were no differences in blood pressure between the sham operation group and the control group. Compared with the control group,both systolic blood pressure and diastolic blood pressure in three SAD groups fluctuated obviously within 24 hours.The level of Ca2+ of mitochondrion was elevated significantly(P<0.01)and mitochondrial membrane potential was decreased significantly(P<0.01)in the three SAD groups compared with that in the control group and sham operation group. And in the three SAD groups,the level of Ca2+ of mitochondrionwas gradual y increased and mitochondrial membrane potential was gradual y decreased(P<0.01). Conclusion The fluctuation of blood pressure caused by sinoaortic denervation,which is a significant damage to the mitochondria of brain cel s,and is becoming more and more serious with time.It is very important to maintain the stability of blood pressure.%目的:探讨去窦弓神经(SAD)对大鼠神经细胞线粒体 Ca2+和膜电位的影响。方法选取 SD大鼠50只随机分为对照组,假手术组,SAD30 d组、SAD60 d组、SAD90 d组,采用火焰原子吸收法测线粒体 Ca2+浓度,激光共聚焦显微镜检测膜电位。结果假手术组血压与对照组比较差异无统计学意义,SAD组与对照组比较24 h舒张压和收缩压波动明显(P<0.01);SAD30 d组、SAD60 d组、SAD90 d组与对照组及假手术组比较 Ca2+浓度明显升高(P<0.01),膜电位明显降低(P<0.01);而且 SAD组随时间延长Ca2+浓度逐渐升高(P<0.01),膜电位逐渐降低(P<0.01)。结论去窦弓神经后导致的血压波动,对脑细胞线粒体造成明显损害,且随时间延长日益严重,因此维持血压稳定至关重要。
