远志总皂苷对癫痫模型大鼠学习记忆及海马LTP的影响

         

摘要

Objective To observe the effects of Tenuigenin (TEN) on the ability of spatial learning and memory and long-term potentiation (LTP) in epilepsy model rats by intraperitoneal injection of Pentylenetetrazole (PTZ) solution.Methods Male wistar rats were randomly divided into control group,epilepsy model group,and tenuigenin group.Morris water maze test was performed to assess the ability of spatial learning and memory of animals.Additionally,electrophysiological technique was used to observe the changes in rat hippocampal LTP through the comparison of field excitatory postsynaptic potential (fEPSP) amplitudes in hippocampal CA1 region before and after high stimulation(HFS).Meanwhile,the regulation effect of TEN on above indices were studies.Results Compared with control group,the ability of spatial learning and memory as well as LTP were obviously decreased in epilepsy model group,however,TEN improved the abnormal changes of above indices.Conclusion TEN can improve the ability of learning and memory abilities and promote in vivo hippocampal LTP.%目的 观察远志总皂苷(TEN)对戊四氮(PTZ)点燃癫痫模型大鼠空间学习记忆能力及海马长时程增强(LTP)的影响.方法 雄性Wister大鼠随机分为对照组、癫痫模型组、TEN组,利用水迷宫法观察各组大鼠的空间学习记忆能力,利用电生理学方法通过对高频刺激前后海马CA1区场兴奋性突触后电位(fEPSP)幅度比较检测大鼠海马LTP变化.结果 与对照组相比较,癫痫模型组空间学习记忆能力显著下降,海马CA1区LTP受到显著抑制(P<0.05);TEN可改善上述指标的异常变化(P<0.05).结论 TEN可改善癫痫模型大鼠空间学习记忆能力及海马LTP的抑制.
