痰瘀清方对自发性高血压大鼠血压及动脉重构的影响

         

摘要

目的 观察痰瘀清方(TYQF)对自发性高血压大鼠(SHR)的降压作用,对SHR胸主动脉和肠系膜上动脉2级分支重构的影响,研究TYQF抗高血压的作用机制.方法 将11周龄雄性SHR 40只随机分为5组:TYQF高、中、低剂量组,福辛普利组和SHR对照组,每组8只.另以8只同周龄雄性WKY大鼠为WKY对照组.TYQF高、中、低剂量组给药剂量分别为23.24 g/kg、11.62 g/kg、5.81 g/kg,福辛普利组给药剂量为0.9 mg/kg,连续给药8周.SHR对照组和WKY对照组给予等容积双蒸水.用药前及用药后每2周测量血压及心率.20周龄时采集胸主动脉和肠系膜上动脉2级分支标本.经HE染色观察胸主动脉和肠系膜上动脉2级分支病理结构变化;经Masson染色观察胸主动脉和肠系膜上动脉2级分支血管中膜胶原容积分数(CVF)的变化.结果 SHR对照组血压较WKY对照组升高(P<0.001);TYQF高、中剂量组大鼠血压较SHR对照组降低(P<0.001).大鼠胸主动脉血管外径(ED)、血管内径(LD)、血管中膜厚度(MT)、血管中膜厚度与内径比(MT/LD)值(P<0.05,P<0.01,P<0.001)与血压均呈正相关.SHR胸主动脉ED、LD、MT、MT/LD值均较WKY对照组增高(P<0.05或P<0.001).TYQF高剂量组ED、LD值较SHR对照组减小(P<0.05或P<0.01);MT值较SHR对照组减小,但差异无统计学意义;MT/LD值较SHR对照组则无明显改变.SHR胸主动脉血管中膜CVF值均较WKY对照组增高(P<0.001);TYQF高、中剂量组均较SHR对照组降低(P<0.05或P<0.001).SHR肠系膜上动脉2级分支ED值较WKY对照组无明显变化,LD值较WKY对照组减小(P<0.01);MT、MT/LD值较WKY对照组增高(P<0.05,P<0.01,P<0.001).TYQF高剂量组LD值较SHR对照组、TYQF中剂量组、TYQF低剂量组增高(P<0.05,P<0.01,P<0.001);MT/LD值较SHR对照组、TYQF中剂量组、TYQF低剂量组减小(P<0.05,P<0.01,P<0.001);其他指标较SHR对照组无明显变化.SHR肠系膜上动脉2级分支血管中膜CVF值较WKY对照组增高(P<0.001);TYQF高、中剂量组较SHR对照组降低(P<0.05);TYQF高、中剂量组较TYQF低剂量组降低(P<0.01).结论 TYQF具有降低SHR血压的作用.TYQF可能对SHR胸主动脉和肠系膜上动脉2级分支的血管重构具有一定的改善作用.%Objective To observe the effects of Tanyuqing formula(TYQF) on the blood pressure,the arterial remodeling of the thoracic aorta and the second branch of superior mesenteric artery in spontaneously hypertensive rat (SHR),and to study the mechanism of antihypertensive effect of TYQF.Methods Forty male SHR were assigned to 5 groups randomly:SHR control group,SHR+Fosinopril,SHR+ TYQF high dose group,SHR+ TYQF medium dose group,SHR+ TYQF low dose group.Each group consisted of 8 animals.8 male WKY rats (11 week-old) were used as normal control group.The rats in SHR+ TYQF high dose group,medium and low dose group were given TYQF contained crude drug of 23.24 g/kg,11.62 g/kg,5.81 g/kg respectively.The rats in SHR+Fosinopril were given suspl of fosinopril of 0.9 mg/kg.The rats in SHR and WKY control group were given the distilled water of the same volume.Drugs were given for 8 weeks constantly.Before treated and after every two weeks,the blood pressure and heart rate of every rat was measured.Thoracic aorta and the second branch of superior mesenteric artery were harvested at 20 week-old.After hematoxylin and eosin staining,the external diameter (ED),lumen diameter (LD),medial thickness (MT),medial thickness/lumen diameter (MT/LD) of aorta and the second branch of superior mesenteric artery were measured.After Masson's trichrome staining,the collagen volume fraction (CVF) of tunica media of aorta and the second branch of superior mesenteric artery were measured.Results There was a significant positive correlation between the blood pressure (P<0.001).The blood pressure was increased compared with that of WKY (P<0.001).The blood pressure in SHR+ TYQF high and medium dose group were reduced compared with that of SHR (P<0.001).The values of ED,LD,MT,MT/LD of thoracic aorta of SHR were increased compared with that of WKY (P<0.05 or P<0.001).The values of ED,LD in SHR+ TYQF high dose group were reduced compared with that of SHR (P<0.05 or P<0.01).The values of MT in SHR+ TYQF high dose group were reduced compared with that of SHR,but there was no statistic significance.The values of MT/LD of SHR were not significantly different from that of three groups of SHR+ TYQF.The values of CVF in SHR were increased compared with that of WKY (P<0.001).The values of CVF of thoracic aorta SHR+ TYQF high and medium dose group were reduced compared with that of SHR (P<0.05 or P<0.001).The values of ED of the second branch of superior mesenteric artery of SHR were not significantly different from that of WKY.The values of LD in SHR were reduced compared with that of WKY (P<0.001).The values of MT,MT/LD in SHR were increased compared with that of WKY (P<0.05,P<0.01,P<0.001).The values of LD in SHR+ TYQP high dose group was increased compared with that of SHR,SHR+ TYQPF medium and low dose group(P<0.05,P<0.01,P<0.001).The values of MT/LD in SHR+ TYQF high dose group was reduced compared with that of SHR,SHR+ TYQF medium and low dose group(P<0.05,P<0.01,P<0.001).Other index of SHR was not significantly different from that of three groups of SHR+ TYQF.The values of CVF of tunica media of the second branch of superior mesenteric artery of SHR were increased compared with that of WKY (P<0.05).The values of SHR+ TYQF high and medium dose group were reduced compared with that of SHR (P<0.05).The values of SHR+ TYQF high and medium dose group were reduced compared with that of SHR+ TYQF low dose group(P<0.01).Conclusion TYQF could decrease the blood pressure and heart rate in SHR.TYQF could improve the remodeling of thoracic aorta and the second branch of superior mesenteric artery in SHR.
