不同剂量碘海醇对大鼠肾脏氧自由基的影响

         

摘要

目的:探讨不同剂量碘海醇对大鼠肾脏氧自由基的影响,为造影剂的安全使用提供依据.方法:将40只Wister大鼠随机分为成4组:空白对照组(A)、小剂量碘海醇组(B)、中剂量碘海醇组(C)、大剂量碘海醇组(D).B、C、D组尾静脉注射碘海醇,24 h后检测血清中肌酐(Scr)水平、肾组织中丙二醛(MDA)含量及超氧化物歧化酶(SOD)活力,显微镜下观察肾脏病理改变.结果:A、B、C、D四组血清中Cr水平分别为(46.21±8.25)、(60.54±13.34)、(80.16±15.26)、(94.27±16.81)μmol/L,肾脏组织MDA含量分别为(2.74±0.48)、(4.51±1.47)、(6.92±2.18)、(8.87±2.31) nmol/mg蛋白,SOD活力分别(195.21±11.01)、(170.24±13.59)、(140.86±25.91)、(121.54±25.50)U/mg蛋白,各组间差异有统计学意义(P<0.05).结论:肾脏内氧自由基随着碘海醇剂量的增加而增加,呈正相关.%Objective:To investigate the effects of different doses of iohexol injected on oxygen free radicals of kidney in rats. Methods:A total of 40 rats were randomly divided into 4 groups: blank control group( A ), small dose of iohexol group( B ), moderate dose of iohexol group( C ), large dose of iohexol group( D ). After 24 h blood samples were collected for measurement of serum creatinine( Cr ) level. The rats were sacrificed and the renal tissues were removed for determination of MDA content and SOD activities. Pathological changes in renal tissues were observed with light microscopes. Results:The concentrations of serum Cr in group A, B, C and D were ( 46.21 ± 8. 25 ),( 60. 54 ± 13. 34 ),( 80. 16 ± 15. 26 ),( 94. 27 ±16.81 )μmol/L. The content of MDA in renal tissues were (2.74 ±0.48 ),( 4. 22 ± 1. 40 ),( 6. 15 ± 1. 88 ),(8.07 ±2.21 )ranol/mg protein, and activities of SOD were ( 195.21 ± 11.01 ),( 175.64 ±13.43 ),( 146.36 ±21. 11 ),( 125.57 ±22.55 )U/mg protein. Difference between the four groups was statistically significant( P <0. 05 ). Conclusion: The oxygen free radicals in the kidney increase with the increase of the dose of iohexol. There are positively correlation between them.
