缬沙坦对早期糖尿病肾病大鼠肾组织TGF-β1/Smad2的影响

         

摘要

目的:探讨血管紧张素Ⅱ受体拮抗剂缬沙坦(代文)对早期糖尿病肾病大鼠肾小管上皮细胞丝/苏氨酸激酶受体(Smad2) 的影响.方法:用代文灌服DN模型大鼠,生化检测实验大鼠24 h尿蛋白、血肌酐(Scr)、尿素氮(BUN)、血浆白蛋白(Alb);PAS染色观察大鼠肾脏病理变化;免疫组化(IHC) 检测肾组织TGF-β1/Smad2表达.结果:缬沙坦能够降低实验大鼠肾小管上皮细胞Smad2的表达,减少24 h尿蛋白、Scr、BUN,升高Alb,缩小肾小球直径,改善肾脏病理,缬沙坦组与模型组相比,P<0.05或P<0.01.结论:缬沙坦可能通过影响DN大鼠肾组织TGF-β1/Smad2的表达改善上述生化指标,从而减轻肾脏病理损害.%Objective: Discuss the impact of Angiotensin II receptor antagonist valsartan ( Diovan ) in renal tubular epithelial cells silk / threonine kinase receptors ( Smad2 ) of early diabetic nephropathy. Methods: With Diovan filling DN model rats, then rats 24 h urine protein, serum creatinine ( Scr ), urea nitrogen ( BUN ), plasma albumin ( Alb ) were detected by biochemical. PAS staining was used to observe the pathological changes of the rat kidney. TGF - β1/Smad2 expression of renal tissue was detected by immunohistochemistry ( IHC ). Results: Valsartan can reduce the expression of Smad2 on experimental rats renal tubular epithelial cells, 24 h urine protein, Scr and BUN, elevated Alb,reduced glomerular diameter, improve renal pathology. Valsartan group compared with the model group, P<0. 05 or P< 0.01. Conclusion: Valsartan may improve the biochemical indicators, by affecting the expression of DN rats kidney TGF - β1/Smad2,thereby reducing renal pathological damage.
