金匮肾气丸对庆大霉素诱导肾损伤大鼠肾组织Notch2/hes1信号通路的影响

         

摘要

Objective: To dynamic observe expression of Notch2/hesl in gentamicin - induced renal injury and repair process and Jinguishenqiwan affect on it. Methods: Seventy - two mature rats were randomly divided into the normal group( saline ga-vage and intraperitoneal injection, once daily,held following the ten days )、the Jinguishenqiwan group ( Jinguishenqiwan gavage, once daily, held following ten days; Gen 120 mg/kg, once daily, intraperitoneal injection, continuous ten days ) and gentamicin group( sa-lin gavage; Gen 120 mg/kg, once daily intraperitoneal injection, continuous ten days ). Observed renal damage by light microscopy. By western - blotting, real-time quantitative PCR, observed expression of the Notch2/hesl at l1428day post - modeling. Results: Renal biopsy showed: at 1/14 day, compared with the normal group, the degree of kidney damage is different in the Jinguishenqiwan group and model group; at 28 day, tubular back to normal in the Jinguishenqiwan group and the model group. Western - blotting , RT - PCR shows: at 1/14 day, compared with the normal group, the level of Notch2/hesl increased significantly in model group and Jinguishenqiwan group ( P <0. 05 ); at 28 day, there is no significant difference( P < 0. 05 ); at 1/14 day, compared with the model group, the level of Notch2hesl decreased significantly in Jinguishenqiwan group ( P <0. 05 ); at 28 day, there is no significant difference( P >0. 05 ). Conclusion :Notch2/hesl signaling pathways may be one of the important role in the gentamicin - induced renal injury and repair process; Jinguishenqiwan may be go through inhibit Notch2/hesl signaling pathways to reduce renal injury and promote tubular epithelial cell repair.%目的:动态观察肾气丸对Notch2/hes1通路在庆大霉素诱导肾损伤及修复过程中表达的影响.方法:72只大鼠随机分成空白组(生理盐水灌胃,6 ml/kg;生理盐水腹腔注射,3 ml/kg,1次/d,持继10 d)、金匮肾气丸组(金匮肾气丸灌胃:0.6 g·kg-1·d-1);Gen,120 mg/kg(3 ml/kg,1次/d,腹腔注射,持继10 d)和模型组(生理盐水灌胃,6 ml/kg;Gen,120 mg/kg,1次/d,腹腔注射,持继10 d).应用光镜观察肾小管损伤情况;应用免疫印迹蛋白、实时定量PCR观察大鼠Notch2/hes1信号通路在造模后1、14、28 d肾脏的动态表达情况.结果:肾脏病理切片显示:在第1、14天,与空白组相比,模型组、肾气丸组造成的肾脏损伤程度有所不同;在第28天,各组肾小管基本恢复正常.免疫印迹蛋白、RT-PCR显示:在第1、14天,与空白组相比,模型组和金匮肾气丸组Notch2/hes1的表达明显增高(P<0.05);在第28天,各组表达差异无统计学意义(P>0.05);在第1、14天,与模型组相比,金匮肾气丸组Notch2/hes1的表达明显减少(P<0.05);在第28天,两组表达差异无统计学意义(P>0.05).结论:Notch2/hes1信号通路可能是庆大霉素诱导肾损伤及修复过程中的重要角色之一,肾气丸可能通过抑制Notch2/hes1信号通路减轻肾脏损伤并促进肾小管上皮细胞修复.
