降压合剂对胰岛素抵抗高血压大鼠作用效应及机制的实验研究

     

摘要

目的:观察降压合剂对胰岛素 抵抗高血压大鼠的作用效应,探讨其作用机制。方法:雄性SD大鼠饲 以高糖高盐饲料制模,然后分组给予不同药物灌胃,观察各组大鼠体重、收缩压(SBP) 、空腹血糖(FBG)、空腹血胰岛素(FINS)、胰岛素敏感性指数(ISI)、红细胞Na+K+ATP酶及血 管紧张素Ⅱ(AgⅡ)的变化。结果:(1)高糖、高盐饲料喂大鼠4周时SBP、 FINS升高,ISI降低(P均<0.01),FBG无变化。(2)8周时,与正常组比较,模型组红细 胞 Na+K+ATP酶活性降低,AgⅡ升高(P均<0.05)。与模 型 组比较,用药各组SBP、FBG、FINS下降,ISI升高(P<0.05或P<0.01);高降压合 剂组 、卡托普利组、降压合剂+卡托普利组红细胞Na+K+ATP 酶活性升高(P均<0.05),卡托普利组AgⅡ降低(P<0.05)。结论:高糖高盐饲料喂养大鼠能产生高血压及胰岛素抵抗。降压合剂能降压、降糖、降血 胰岛素 、增加胰岛 素敏感性。降压合剂的降压及增加胰岛素敏感性的作用可能是通过增高Na+ K +ATP酶活性这一途径。%Objective:To observe the effects of Jiangya mixture(JYM,降压合剂) on insulin resistance hy pertensive rat model and investigate its mechanism.Methods:Male Spraque Darley rats were fed on high glucos e and high salt diet to establish experimental model,following by giving differe nt drugs to divided groups.The changes in body weig ht(BW),systolic blood pressure(SBP),fasting blood glucose(FBG),fasting blood ins ulin(FINS),insulin sensitivity index(ISI),red blood cell(RBC) Na+ K+ATPase,angiotensinⅡ(AgⅡ) were observed.Results:(1)At fourth week,SBP and FINS were increased but ISI decrea sed in rats fed on high glucose and high salt diet(all P<0.01).(2)At ei ghth week in comparison with normal group the activity of RBC Na+ K+ATPase was decreased but AgⅡ increased (all P<0.05)in model gro up.In Comparison with model group the SBP,FBG and FIN S were decreased,otherwise ISI was increased (P<0.05 or P<0.01).In o ther groups,meanwhile the activity of RBC Na+K+ATP JYM+capto pril group and AgⅡ decreased (P<0.05) in captop ril group.Conclusions:Hypertension and insulin resistance can be ind uced in rats by feeding high glucose and high salt diet .The JYM can reduce blood pressure,blood sugar,and level of blood insulin other wise increase insulin sensitivity.The mechanisms of it on reducing blood pressur e and increasing insulin sensitivity may be obtained by heightening the activit ies of Na+K+ATPase.
