淫羊藿苷治疗去势大鼠骨质疏松症基因表达谱分析

     

摘要

目的 利用基因芯片技术探究淫羊藿苷治疗去势大鼠骨质疏松症的分子机制.方法 将20只SD雌性大鼠随机分为模型组和治疗组,每组10只,采用去卵巢法造模,治疗组予淫羊藿苷[水相0.4 mL/(100 g·d),油相0.6mL/(100g·d)]灌胃,模型组每日灌胃相应体积冷开水和油中王牌植物油,每日1次,连续1 3周,应用基因芯片技术检测淫羊藿苷治疗后去势骨质疏松症大鼠与模型骨质疏松症大鼠相关基因的表达,通过Gene Ontology数据库对差异表达基因进行基因信息学分析.结果 淫羊藿苷治疗去势大鼠骨质疏松症共有532条基因发生了差异表达,其中有175条基因上调,357条基因下调.结论 多种基因差异表达是淫羊藿苷治疗去势大鼠骨质疏松症的可能分子机制.%Objective To investigate the mechanism of icariin in treating osteoporosis of ovariectomized with gene chips technology. Method A total of 20 female SD rats were randomly divided into model group and treatment group, 10 rats in each group. After ovariectomized to make model, treatment group was given icariin [aqueous phase 0.4 mL/(100 g · d), oil phase 0.6 mL/(100 g · d)J, model group was given the same volume water and vegetable oil once per day for 13 weeks. Gene chips of RatRef-12-v 15590097019 were used to investigate the gene expression profile of osteoporosis rats, and the different expressed genes were analyzed with Gene Ontology. Results A total of 532 significant differentially expressed genes related were detected. Among them, 175 genes were up-regulated, 357 genes were down-regulated. Conclusion Many genes differential expression are related with icariin in treating osteoporosis of ovariectomized rats.
