0bJ3ctive:To study 1he characteristics of CD4 T cells TCR VfS gene cbnning in hepatitisB vaccinated and 1he difference of TCR Vβ genes m onocbnal changes beteeen 1he cbne of genes in respondeis and non responders 1o hepatitis B vaccine .M e1hods:CD4+ T cells of 80 hepatitisB vaccinated individualV(3 genes CDR3 regbn fragmentswere amplified by multi-prmerPCR empUScation m eihods,PCR products w ere aplied on agarose gel etectophoresis and gene scanning by sequencing instnm ent.R esulte :From 80 cases of hepatitis B vaccinat-ed indiridual,58 cases were positive fbrantrHBs and 22 cases were negative .M onocbnal TCR VfS gene changes w ere m ain^r concentrated in V(& ,V(I ,V(I ,V(li ,and V(l7 fim % of genes .The frequency of changes in 1hese fire fen % of genes was significant^ bwer in nonrespondeis group 1han responders (P<0 .01 or P<0 .05) . Condusbn :CD4+ T cells of TCR V(3 gene change is one of 1he in portant iactir 1hat affect 1he body's inmune response to hepatitis B vaccine.%目的:研究乙肝疫苗接种者CD4+T细胞TCR Vβ基因克隆化特征,分析乙肝疫苗有应答者和无应答者TCR Vβ基因单克隆改变的差异性.方法:采用多引物PCR技术扩增80例乙肝疫苗接种者CD4+T细胞TCR Vβ基因22个家族的CDR3区基因片段,对PCR产物分别应用琼脂糖凝胶电泳和基因扫描两种方法检测.结果:80例乙肝疫苗接种者中有58例产生抗体,有22例未产生抗体.TCR Vβ基因单克隆改变主要集中在Vβ2、Vβ8、Vβ9、Vβ11和Vβ17五个家族上,乙肝疫苗无应答组这五个Vβ基因单克隆改变频率明显低于乙肝疫苗有应答组(P<0.01或P<0.05).结论:CD4+T细胞TCR Vβ基因克隆化改变是影响机体对乙肝疫苗免疫应答效果的重要因素之一.
