小胶质细胞在不同损伤神经环境下CD200R的表达

     

摘要

目的:小胶质细胞在不同的损伤神经环境中接受刺激后观察其CD200R的表达情况,探讨活化的小胶质细胞与神经元之间的相互作用.方法:分别采用鼠小胶质细胞瘤细胞BV2细胞和鼠嗜铬细胞瘤细胞PC12细胞进行研究.将BV2细胞分别与正常的及用鱼腾酮损伤的PC12细胞共育培养.MTT检测细胞活力,倒置荧光显微镜观察细胞CD200R表达,ELISA定量检测CD200R蛋白表达.结果:BV接受损伤PC12上清孵育后其CD200R表达显著下降(P<0.05),而与损伤PC12共育后其CD200R表达显著升高(P<0.05).结论:损伤神经细胞上清能够降低小胶质细胞CD200R 的表达,而与损伤神经细胞共育能提高小胶质细胞的CD200R表达.%Objective:To explore the interactions between microglial and neurons by investigating the CD 200R expression on microglial cells in the microenvironment with various conditions of injured neurons . Methods: The rat microglial cells BV2 and rat pheochromocytoma cells PC 12 were used . BV2 were incubated with PC 12 injured with Rotenone in different conditions. Immunofluo— rescence and ELISA was used to demonstrate the CD 200R expression on cells, the quantification of CD200R expression was determined with Cell-based ELISA. Results: Compared with control the CD 200 R expression on BV2 incubated in supernatant of injured PC 12 was reduced significantly (P <0. 05) , while the CD200R expression on BV2 cocultured with injued PC12 was improved (P <0. 05). Conclusion: The supernatant of injured neurons reduces the CD 200R expression, and coculturing with injured PC 12 improves CD200R on microglia.
