苏铁总黄酮对Lewis肺癌模型小鼠VEGF、bFGF、HIF-1α、NF-κB表达的影响

     

摘要

目的:探讨苏铁总黄酮对Lewis肺癌模型小鼠血管内皮细胞生长因子(VEGF)、碱性成纤维细胞生长因子(bFGF)、低氧诱导因子-1α (HIF-1α)、核转录因子(NF-κB)表达的影响.方法:采用HE染色法观察肿瘤组织的病理形态变化,免疫组化法和Western blot法检测肿瘤组织中VEGF、bFGF、HIF-1α、NF-κB蛋白的表达,荧光定量PCR法检测肿瘤组织中VEGF、bFGF、HIF-1α、NF-κBmRNA的表达.结果:免疫组化法、Western blot法和荧光定量PCR法的检测结果显示,三者结果基本一致,与模型组比较,苏铁总黄酮各给药组VEGF、bFGF、HIF-1α、NF-κB基因表达量和对VEGF、bFGF、HIF-1α、NF-κB蛋白表达表达率均降低,差异具有显著性(P<0.05).结论:苏铁总黄酮治疗肺癌的作用机制可能为通过抑制侵袭转移相关基因VEGF、bFGF、HIF-1α、NF-κB的表达,进一步抑制侵袭转移相关功能蛋白VEGF、bFGF、HIF-1α、NF-κB的表达,从而发挥抗肺癌侵袭转移作用.%Objective:To investigate the effects of total flavonoids from Cycas Revolute on expression of vascular endothelial growth factor (VEGF),basic fibroblast growth factor (bFGF),Hypoxia inducible factor-1α (HIF-1α) and nuclear factor-κB(NF-κB) in model mice of Lewis lung cancer.Methods: The expressions of VEGF,bFGF,HIF-1α and NF-κB in tumor tissues were detected by immunohistochemistry and Western blot.The expression of VEGF,VEGF and NF-κB in tumor tissues were detected by fluorescent quantitative PCR.BFGF,HIF-1α and NF-κB mRNA were detected by immunohistochemistry.Results: The results of immunohis-tochemistry,Western blot and Real-time PCR showed that the results were basically the same,compared with model group,the expression of VEGF,bFGF,HIF-1α,NF-κB mRNA and the expression of VEGF,bFGF,HIF-1α and NF-κB were decreased,the difference was highly significant (P<0.05).Conclusion: The mechanism of total flavonoids from Cycas Revolute in the treatment of lung cancer may be through inhibition of the expression of VEGF,bFGF,HIF-1α,NF-κB in invasion and metastasis,and further inhibit the expression of VEGF,bFGF,HIF-1α and NF-κB in invasion and metastasis-related proteins,thus play a role of anti-lung cancer invasion and metastasis.
